牛磺酸对乙醇醛细胞毒性的抑制作用

摘    要:目的:研究牛磺酸对乙醇醛所致细胞毒性的抑制作用。 方法:采用单细胞凝胶电泳技术(彗星实验)检测牛磺酸(50 mmol•L-1)对乙醇醛(GA, 0.1 mmol•L-1)所致小鼠脾淋巴细胞DNA损伤的抑制作用,并用2,4-二硝基苯肼法定量检测牛磺酸对反应体系中乙醇醛含量的影响,同时设乙醇醛阳性对照组和PBS阴性对照组。 结果:在0.1 mmol•L-1乙醇醛溶液中加入50 mmol•L-1牛磺酸后,小鼠脾淋巴细胞DNA的损伤率较阳性对照组明显降低(P<0.01),彗星实验的迁移距离(尾长)也显著缩短(P<0.01),牛磺酸呈剂量依赖性明显降低反应体系中乙醇醛的含量。 结论:牛磺酸对乙醇醛的细胞毒性有明显抑制作用。

Abstract:Objective To investigate the inhibitory effects of Taurine on the cellular toxicity induced by glycolaldehyde. Methods The inhibitory effects of Taurine (50 mmol·L~ -1) on glycolaldehyde (0.1 mmol·L~ -1) induced DNA damage in spleen lymphocytes of mice were studied in vitro using single cell gel electrophoresis (comet assay),and 2,4-dinitrophenylhydrazine (DNPH) measurement was used to detect quantitatively the effects of Taurine on carbonyl compound contents in a reactive system. Results After 50 mmol·L~ -1 Taurine was added to the spleen lymphocyte suspension containing 0.1 mmol·L~ -1 glycolaldehyde, the cellular DNA damage rates were markedly decreased (P<0.01) and the migration distances of DNA were significantly reduced (P<0.01) as compared with positive control group, and Taurine reduced the carbonyl compound contents in a reactive system in a dose-dependent manner. Conclusion Taurine can significantly inhibit the cellular toxicity induced by glycolaldehyde.
