摘    要:目的研究BRG1在乳腺癌组织中的表达,探讨其临床意义及判断预后的价值。方法应用组织芯片和免疫组化方法评估BRG1在437例乳腺癌组织中的表达水平,并分析其与临床病理特征及患者预后的关系。结果在437例乳腺癌组织中,BRG1阴性表达率和阳性表达率分别为47.6%和52.4%;BRG1与年龄、肿瘤大小、淋巴结转移、病理学分级、病理类型、雌激素受体(ER)、孕激素受体(PR)及人类表皮生长因子受体2(HER2)均无明显相关性,差异无统计学意义(P〉0.05);BRG1高表达提示患者5年总生存率及无瘤生存率低(P=0.000,P=0.000)。结论BRG1高表达与乳腺癌的不良预后密切相关,BRG1可能成为判断乳腺癌预后的一个重要标记物。

Abstract:Objective To investigate the expression of BRG1 in breast cancer tissues so as to discuss the role of BRG1 in breast cancer pathogenesis and its clinicopathological significance. Methods The expression of BRG1 was de- tected by immunohistochemistry using a tissue microarray containing 437 specimens of breast cancer patients. The rela- tionship between the levels of BRG1 expression, clinicopathologic parameters, and survival time of the patients were ana- lyzed. Results Of the 437 breast cancer specimens analyzed, low expression levels ( negative and weak) and high ex- pression levels ( moderate and strong) were 47.6% and 52.4% , respectively. The Kaplan - Meier analysis and log - rank test revealed that increased BRG1 expression was associated with poor overall or disease -specific 5 -year patient survival (P =0. 000 and P =0. 000, respectively). However, no significant correlations were found between BRG1 ex- pression and clinicopathological variables that included patient age, tumor size, lymph node metastasis, histology grade, histology type, estrogen receptor (ER) status, progesterone receptor (PR) status, and human epidermal growth factor receptor 2 (HER2) status. Conclusions High BRG1 expression is an independent negative prognostic factor for breast canter patients. BRG1 may be a prognostic marker for breast cancer.
