纳洛酮对心肺复苏后大鼠血浆内皮素水平的影响

摘    要:
目的:探讨纳洛酮对大鼠心肺复苏后血浆内皮素水平的影响。方法:将Wistar雄性大鼠75只随机分为3组:对照组(C组)、肾上腺素组(E组)、纳洛酮组(N组),每组25只。采用窒息致心跳骤停,心跳骤停10min时,经股动脉注入由窒息前采集的动脉血与肝素50U/100g、肾上腺素4μg/100g(E组)或肾上腺素4μg/100g+纳洛酮0.1mg/100g(N组)的混合液进行复苏。对照组不予采血、窒息、复苏。分别于复苏后30、60、120、180min时,采血检测ET含量。结果:复苏后各时点E组、N组血浆ET值明显高于基础值(P<0.05),且E组血浆ET值随时间进行性增加;复苏后120min、180min,N组血浆ET值明显低于E组(P<0.05)。结论:心肺复苏时应用纳洛酮可降低血浆内皮素水平,有利于复苏后心脑功能的恢复。

Abstract:
Objective :To evaluate the effect of Naloxone given during cardiopulmonary resuscitation(CPR) on plasma endothelin(ET).Methods: Seventy-five male Wistar rats were randomly divided into 3 equal groups of 25 animals: control group(group C);epinephrine group(group E) and naloxone group(group N).Cardiac arrest was induced by asphyxia in group E and N while group C was subjected to no asphyxia.CPR was started at 10 min of cardiac arrest and heparin 50U/100g,epinephrine 4μg/100g or epinephrine 4μg/100g+ Naloxone 0.1 mg/100 g were given via femoral artery.Blood samples were taken from internal jungular vein at 30,60,120 and 180 min after successful resuscitation respectively for determination of plasma ET content.Results :The values of ET were significantly higher than baseline in groups E and N after resuscitation(P<0.05).The values of ET in group E increased with the prolongation of resuscitation.The values of ET in group N were significantly lower than those in group E 120 and 180 min after resuscitation(P<0.05).Conclusion: Naloxone can decrease the level of ET in plasma,which may be advantageous to the recovery of cardiac and cerebral function after resuscitation.
