LA795小鼠肺腺癌细胞系不同肺转移率克隆株流变特性的研究

摘    要:
本文对 LA795小鼠肺腺癌细胞系进行克隆分离,并以不同克隆株接种于 T739同基因小鼠,从而得到非转移、低转移及中转移三个瘤株,而后测定其细胞电泳时间、膜脂流动性、粘附性及剪切应力耐受性。结果表明:转移瘤株的细胞电泳率及粘附性均高于非转移瘤株,三个瘤株膜脂流动性与其肺转移率呈负相关关系,中转移瘤株剪切应力耐受性低于非转移及低转移瘤株。结合我们过去对不同转移率及途径瘤株接种于实验动物后引起其血液宏观流变性变化的研究,我们认为:肿瘤侵袭、转移过程与血液、血细胞及肿瘤自身细胞流变特性变化密切相关,在肿瘤发展早期提高血液及血细胞流变性,而在晚期降低其流变性,对抑制肿瘤转移有一定意义。

Abstract:
The cells of mouse lung adenocarcinoma LA 795 were divided into 15 monocolony lines. That the cell electrophoresis time (EPT) of the 15 lines were measured, among which five have significant dif- ference on the EPT) were selected. The five selected cell lines were respectively transplanted into T739 -strain inbred mice. These mice were then killied and their lung metastasis rates were examined on the 20th day after the transplantation of tumor. Through observing the lung metastasis rates, the non -metastatic (LA-1), low-metastatic (LA-D) and mid-metastatic (LA-5) tumor cell lines were obtained among the five. The following indexes, cell elcctrophoresis time (EPT), membrane fluidity, adhesion and endurance to shear stress were measured for LA-1, LA - D and LA -5. The results in -dicated 1) The cells of metastatic lines (LA - D and LA-5) have a shorter electrophoresis time and higher adhesion compared with non -metastatic line (LA-1). 2) The membrane fluidity of the three cell lines is negatively correlated with their lung metastatic rates. 3) The cell survival rate of the mid -metastatic line LA-5 was lower than that of the non-metastatic line LA-D and low -metastatic line LA- 5 on fixed action time of shear stress. These results show that there is a close relationship be- tween rheological properties of tumor cells and their invasion and metastasis. The results also imply that the increase of the blood rheological properties will raise the shear stress of blood flow , which is beneficial to kill tumor cells, and therefore may help to inhibit the metastasis of tumor.
