蜂胶总黄酮对全脑缺血再灌注大鼠脑损伤的保护作用

摘    要:目的探讨蜂胶总黄酮(TFP)对全脑缺血再灌注大鼠脑组织损伤的保护作用并对其机制进行探讨。方法采用48只清洁级雄性SD大鼠,随机分为假手术组、模型组、灯盏花素(50 mg/kg)对照组和TFP低、中、高剂量(25,50,100 mg/kg)组,模型组和用药组采用四血管阻断法制备全脑缺血再灌注模型。分别观察低、中、高剂量的TFP对全脑缺血再灌注后大鼠脑神经功能(NSS评分)、脑梗死面积、脑组织内超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量的影响。结果模型组NSS评分、脑梗死面积、脑组织内SOD活力和MDA含量与假手术组比较均有统计学意义(P〈0.01)。TFP低、中、高剂量组均能使全脑缺血再灌注大鼠NSS评分明显降低,与模型组比较均具有统计学意义(P〈0.01);TFP中、高剂量组能使大鼠脑梗死面积和脑组织内MDA含量明显降低,与模型组比较有统计学意义(P〈0.01);能使SOD含量显著升高,与模型组比较差异亦具有统计学意义(P〈0.01)。结论 TFP能显著改善大鼠全脑缺血再灌注后脑神经功能,使脑组织梗死面积与MDA含量显著降低、SOD活力显著升高,能减轻缺血再灌注对大鼠脑组织的损害,具有一定的脑保护作用。

Abstract:Objeetlve To explore the protection effect and mechanism of the total flavonoids of propolis(TFP) on the cere- bral ischemia and reperfusion of brain injury in rats. Methods The 48 SD male rats of clean agent were randomly divided into sham operated group, model group, erigeron(50 mg/kg) group, TFP low, middle and high dose(25,50,100 mg/kg) groups. Pullsinelli - 4 vessel occlusion method was used by the model group and the drug group to prepare cerebral ischemia reperfusion injury model. The effects of TFP of low, middle and high dose on brain neural of rats with cerebral isehemia and reperfusion (NSS scores), cerebral ischemia area, activity of SOD and content of MDA in the brain were measured in each group. Results Compared to the sham operated group, the NSS scores, cerebral ischemia area, activity of SOD and content of MDA in the brain in the model group had statistical significance( P 〈 0.01 ). Compared with the model group, TFP three dose groups can significantly decrease the NSS scores of rats with cerebral ischemia and reperfusion, which had statistical significance( P 〈 0.01 ). Compared with the model group, TFP middle and high dose group can obviously decrease the cere- bral ischemic area and the content of MDA in the brain, which had statistical significance( P 〈 0.01 ). And compared with the model group, it can increase the activity of SOD, which also had statistical significance ( P 〈 0.01 ). Conclusion TFP can significantly improve brain neural of rats with cerebral ischemia and reperfusion, decrease the cerebral ischemic area and the content of MDA in the brain, increase the activity of SOD and ease the impairment of cerebral ischemia and reperfusion on brain tissue of rats. It has a protective effect on the brain.
