一氧化氮在急性出血坏死性胰腺炎大鼠胰肾损伤中的作用

摘    要:目的 探讨一氧化氮(NO)在不同时机及剂量条件下对急性出血坏死性胰腺炎(AHNP)胰、肾损伤的作用。方法 观察不同时机、不同剂量的L-精氨酸(L-Arg)催化NO生成后的AHNP大鼠血浆淀粉酶、肌酐、尿素氮、内毒素、NO,胰、肾组织及NO及血流变化,并分析动物死亡情况。结果 小剂量L-Arg即时给予明显降低AHNP大鼠血淀粉酶,改善胰、肾血循环、保护肾功能、降低大鼠死亡率,延时给予对AHNP病情无影响,大剂量L-Arg无论即时或延时给予,明显加重胰、肾血循环紊乱,肾功能明显受损,大鼠死亡率明显增加。结论 AHNP大鼠产生的NO对胰、肾组织具有截然不同 的生物学作用,这种作用与NO产生明显存在时机与量的关系,对于临床应用NO供体治疗AHNP具有重要意义。

Abstract:Objective To determine the role of different doses and time of nitric oxide (NO) on the pancreas and kidney organ damage in rats with acute hemorrhagic necrotizing pancreatitis (AHNP). Methods After the model of AHNP was established by injection of 5% sodium taurocholate into the biliary pancreatic duct in rats, the changes of amylase, creatinine, ureanitrogen, endotoxin, No in blood and NO in pancreatic and kidney tissue and blood flow of pancreas and kidney were observed in AHNP model treated by different doses and time of L arginine, and the mortality was analyzed. Results Concertration of amylase and the mortality was decreased, blood flow of pancreas and kidney was improve in AHNP model treated immediately by low doses of L arginine but high doses of L arginine could induced severe pancreatic and renal histolcagic damage and the mortatity was incereased. Conclusions Effect of NO in the pathogenesis of pancreas and kidney organ damage in rats with AHNP which is positively correlated with time and doses.
