复方三根制剂配合瘤内注射DDP调控荷肺癌小鼠Bcl-2、P53基因表达的实验研究

摘    要:目的:观察与探讨Bcl-2及P53基因在荷肺癌小鼠经复方三根制剂配合瘤内注射治疗后的表达情况。方法:用lewis肺癌瘤株造模于38只C57BL/6近交系小鼠右腋下,8天后将其随机分成2组:A组(13只)、B组(12只)、C组(13只),并分别予以A组:复方三根制剂加瘤内注射DDP治疗,B组:单纯DDP瘤内注射治疗,C组:瘤内注射NS对照。21天后处死所有小鼠并取下肿瘤标本作HE染色及免疫组化染色检测,观察Bcl-2、P53表达情况。结果:Bcl-2表达情况:A组为46.1%,B组为58.3%,C组为61.5%;P53表达情况:A组为38.4%,B组为41.67%,C组69.2%。结论:瘤内注射DDP可以通过降低Bcl-2、MTP53表达途径促进肿瘤细胞凋亡,而中药复方三根制荆能协同DDP进一步下调两者的表达。

Abstract:Objective:To observe the statement of gene Bcl-2 and P53 in mice bearing lewis lung cancer after administered DDP by intratumor injection and FFSG mixture by oral.Methods:To prepare model mice of c57b1/6 planted lewis lung cancer.The models were randomly divided into three groups, marked group A, group B and C in 8 days.Two groups were treated with DDP by intratumor injection and FFSG mixture by oral and simply administered DDP by intratumor injection respectively.Group C was administered DDP by intratumor injection as control.After administered 21 days, all mice were killed and the samples of tumor were removed.The state of gene Bcl-2 and P53 in samples was measured by immunology history biology method.Results:The positive rates of states of Bcl-2 are 46.1%(A), 58.3%(B) and 61.5% (C) respectively .There are significant differences between A and C. The positive rates of states of P53 are 38.4%(A), 41.67%(B) and 69.2%(C) respectively.There are also significant differences between A or B and C. Conclusion:DDP by intratumor injection may promote tumor apoptosis through regulating down Bcl-2 and P53 pathway,and these functions can be facilitated by a FFSG mixture.
