十全大补汤多糖成分抑瘤及免疫调节作用的初步研究

摘    要:目的:研究十全大补汤总多糖抗肿瘤及对脾细胞功能的调节作用。方法:40只昆明种小鼠,以皮下接种H22细胞的方法制备荷瘤小鼠模型,24 h后随机分为5组,即阴性对照组、顺铂(1 mg/kg)化疗组、顺铂联合高(168.4 mg/m L)、中(84.2 mg/m L)、低(42.1 mg/m L)剂量十全大补汤总多糖组。十全大补汤总多糖采取灌胃给药,0.5 m L/只,1次/d,10 d后眼球放血处死小鼠,称取体质量和瘤质量,计算抑瘤率;分离血清,并分别制备肿瘤细胞和脾细胞悬液,流式细胞仪检测肿瘤细胞和脾细胞的凋亡;ELISA法检测血清中IL-2,IFN-γ和TNF-α含量。结果:顺铂联合各剂量十全大补汤总多糖组小鼠体质量均高于单纯顺铂组,而中、高剂量多糖组的瘤质量均低于单纯顺铂组;顺铂联合各剂量十全大补汤总多糖组肿瘤细胞的凋亡率明显高于单纯顺铂组,而脾细胞的凋亡率均明显又低于单纯顺铂组(P〈0.05);顺铂联合各剂量十全大补汤总多糖组IL-2,IFN-γ和TNF-α的分泌水平均明显高于单纯顺铂组(P〈0.05)。结论:十全大补汤总多糖可促进顺铂诱导肿瘤细胞凋亡,并能保护顺铂对脾细胞的损伤作用、促进荷瘤小鼠IL-2,IFN-γ和TNF-α的分泌,与顺铂联合应用具有减毒增效作用。

Preliminary Study on Polysaccharides of Shiquan Dabu Tang in Resisting Tumor and Regulating Immune

Abstract:Objective: to study the total polysaccharides of Shiquan Dabu Tang in resisting tumor and regulating spleen cells functions. Methods: 40 tumor-bearing mice,prepared with hypodermic inoculation of H22 cell for 24 hours,were randomly divided into five groups: negative control group,chemotherapy with Cisplatin group( 1mg / kg),high dosage of total polysaccharides of Shiquan Dabu Tang combined with Cisplatin group( 168. 4 mg / m L), medium dosage group( 84. 2 mg / m L) and low dosage group( 42. 1 mg/m L). Shiquan Dabu Tang was given through intragastric administration,0. 5 m L per mouse,once a day. 10 days later the mice were killed by eyeball bloodletting. Weigh the body and the tumor,count the rate of resisting tumor; separate blood serum and prepare tumor cell and spleen cell suspension respectively; detect the apoptosis of tumor cell and spleen cell with flow cytometer; detect IL-2,IFN-γ and TNF-α contents in serum with ELISA. Results: compared to Cisplatin group,the weights of mice of all dosage groups were higher,while the weights of tumors of high and medium dosage groups were lower; the apoptosis rates of tumor cells of all dosage groups were higher while the apoptosis rates of spleen cells were lower( P〈0. 05); IL-2,IFN-γ and TNF-α of all dosage groups were higher( P〈0. 05),all significantly. Conclusion: the total polysaccharides of Shiquan Dabu Tang can facilitate the apoptosis of tumor cells induced by Cisplatin,protect the spleen cells damaged by Cisplatin,and improve the excretion of IL-2,IFN-γ and TNF-α in the tumor-bearing mice. It is able to reduce the toxin and increase the effectiveness if combined with Cisplatin.
