牛血小板衍化生长因子对人牙周膜成纤维细胞的生物学效应

摘    要:
目的:观察在牛血小板衍化生长因子作用下人牙周膜成纤维细胞DNA和胶原蛋白合成的情况。方法:采用体外细胞培养技术和核素掺入法。结果:20ng/ml~60ng/ml牛血小板衍化生长因子可明显促进人牙周膜成纤维细胞DNA合成,40ng/ml浓度使细胞DNA合成在24h达最高峰;对细胞的胶原蛋白合成无明显促进作用。结论:牛血小板衍化生长因子对人牙周膜成纤维细胞DNA合成有促进作用,在牙周组织的创伤修复中可能起重要作用。

Abstract:
Aim: DNA and collegen protein synthesis of human periodontal ligament fibroblast(HPLF) were determined by treating with bovine platelet-derived growth factor (bPDGF). Methods: 3H-TdR and 3H-proline incorporation in vitro. Results: bPDGF significantly accelerated DNA synthesis of HPLF in dose-dependent manner over a concertration range of 20ng/ml to 60ng/ml, with a maximal response at a concentration of 40ng/ml at 24h. bPDGF did not stimulate the collegen protein synthesis of HPLF. Conclusions: bPDGF could promote DNA synthesis of HPLF dependently, and may play an important role in wound healing of periodontal tissue.
