MXR7基因在人肝癌组织中的表达及其临床意义

 目的 检测MXR7基因在人肝细胞癌组织中的表达.方法 应用32P标记的MXR7cDNA作为探针,通过Northern blot分析对正常人12种不同组织和病理证实的术前未做治疗的7例非肝肿瘤组织及30例肝癌和癌旁肝组织、12例正常肝组织中MXR7基因的表达进行检测.结果 MXR7mRNA在人肝脏等12种正常组织及7例其它部位肿瘤组织中均不表达;在人肝癌中呈高表达,而癌旁肝组织中呈低表达,其阳性表达率分别为76.7%和13.3%.在血清AFP阴性的肝癌患者和肿瘤直径<5 cm的肝癌中,MXRTmRNA阳性表达率分别为90%和83.3%.肝癌患者手术后1 a和2 a生存率分别是59.1%和54.5%.结论 MXR7基因在人肝癌组织中的高水平表达具有普遍性、特异性和早期性,提示MXR7基因可以作为肝癌的生物学标志物,对肝穿刺组织进行MXR7基因表达检测有助于发现AFP阴性的早期肝癌.未发现MXR7mRNA阳性表达与肝癌患者的预后有明确关系.

Expression of MXR7mRNA in human hepatocellular carcinoma and its clinic significance

Objective To study the expression of MXR7mRNA in human hepatocellular carcinoma and its clinic significance.Methods The 32 P-labeled and purified PCR product of MXR7cDNA wasused as a probe.MXR7mRNA expression was detected by Northern blot analysis in 12 different human normal tissues,7 preoperatively untreated non-liver tumor tissues,30 preoperatively untreated HCC and the corresponding surrounding non-cancerous hepatic tissues samples ,12 normal liver tissue confirmed pathologically.Results There were no detectable expressions of MXR7mRNA in 12 human normal tissues including liver,7 non-HCC tumor tissues and 12 normal liver tissuesmand the frequencies of MXR7mRNA expression in HCC and the corresponding surrounding non-cancerous hepatic tissues were 76.6% and 13.3%,respectivelies.The frequency of MXR7mRNA expression in HCC without serum AFP elevation and in HCC<5 cm were 90%(9/10) and 83.3%(5/6),respectively.The postoperative survival rates for 1 year and 2 years were 59.1% and 54.5%,respectively.Conclusions High expression of MXR7mRNA in human HCC is common and specific and MXR7mRNA may be expressed at early stage of HCC,suggesting that MXR7mRNA may serve as tumor biomarker for HCC.The detection of MXR7mRNA expression in liver puncture tissues is valuable for finding small HCC without serum AFP elevation in the subclinical stage.Our data have not evidenced correlation of MXR7mRNA expression correlated with prognosis of patient
