喉癌组织中P16的免疫组化定量分析

摘    要:目的 :探讨 P1 6在喉癌组织中的表达及和喉癌发生发展的关系。方法 :采用免疫组化S- P法 ,对 56例喉癌组织 ,30例喉癌旁组织及 1 0例正常喉粘膜中 P1 6的表达进行检测 ,应用全自动图像分析仪对染色结果进行定量测定。结果 :喉癌组织中 P1 6的表达量明显低于癌旁组织及正常喉粘膜 ( P<0 .0 5) ,喉鳞状细胞癌 级明显低于 ~ 级 ( P<0 .0 5) ,颈淋巴结转移组明显低于无颈淋巴结转移组 ( P<0 .0 5)。结论 :P1 6在喉癌组织中的表达常为缺失 ,说明 P1 6作为抑癌基因在喉癌的发生中起作用。 P1 6的表达程度越低 ,分化越差 ,越易于浸润和转移 ,P1 6的检测在临床上可能作为估计预后的指标之一。

Abstract:Objective:To study the function of P16 in laryngocarcinoma cell proliferation cycle and the relation between P16 and the genesis and progress of laryngocarcinoma.Methods:Immunohistochemical S-P method was applied to examine the expression of P16 in 56 laryngocarcinomas,30 pericancerous mucosas and 10 normal laryngeal mucosas and the image analysis technology was applied to measure the intensity of staining quantitatively.Results:P16 expression quantity in laryngocarcinomas was lower than that in pericancerous mucosas and normal laryngeal mucosas apparently (P<005).We also studied differentiation and metastasis of laryngocarcinoma.The results showed that P16 expression quantity in larygeal squasmous cell carcinoma grade Ⅲ was lower than that in grade Ⅱ and grade Ⅰ apparently (P<005).It was much lower in laryngocarcinomas with cervical lymph node metastasis than that in laryngocarcinomas without cevical lymph node metastasis (P<005). Conclusion:P16 always displays deletion in laryngocarcinoma,and P16 acting as a tumor suppressor gene plays an important role in laryngocarcinogenesis.The laryngocarcinomas with lower P16 expression represent a strong proliferation ability,poor differentation,then they are easy to infiltrate and metastasize and have unfavorable prognosis.P16 status might become a parameter for evaluating prognosis. 〔
