汉滩病毒核酸疫苗滴鼻免疫小鼠效果的实验

摘    要:目的 观察汉滩病毒DNA疫苗滴鼻免疫诱导机体产生的免疫应答 ,探索汉滩病毒DNA疫苗新的免疫途径。方法 用重组质粒PcDNA3 1B -S1 3 经滴鼻接种BALB/c小鼠 ,采用ELISA、淋巴细胞转化试验及流式细胞仪等方法检测了其诱导的系统和粘膜免疫反应。结果 免疫小鼠血清IgG及粪便IgA明显增高 (P <0 0 1) ,且IgA增幅较大 ;实验组淋巴细胞增殖反应明显 ,刺激指数 (SI)显著增大 (P <0 0 5 ) ;免疫后CD+ 4 、CD+ 8T细胞均增加 (P <0 0 1) ,而CD+ 4 /CD+ 8T细胞比值无显著变化 (P >0 0 5 )。结论 汉滩病毒重组质粒滴鼻免疫能诱导机体产生了特异的系统免疫和较强的粘膜免疫 ,滴鼻的疫苗免疫途径有着潜在的应用价值。

Abstract:To investigate the immune responses induced by intranasal immunization with DNA vaccine of Hantaan virus and to search for a new means of inoculation,BALB/c mice were inoculated intranasally with the recombinant plasmid pcDNA3.1B-S 1.3 expressing nucleocapsid protein gene of Hantaan virus ,and the mucosal and systemic immune responses induced by the DNA vaccine were determined by ELISA.Lymphocyle transformation test and flow cytometry analysis (FACS).It was found that the levels of IgG in serum and IgA in feces in the experimental group of mice were significantly elevated;and the stimulation index (ST) of lymphocytes of the experimental group was higher than that of the control group.After immunization,the numbers of CD 4 and CD 8 T cells were all increased,but the CD 4 T cells/CD 8 T cells ration showed little changes.It concludes from the above experiment that the intranasal immunization of BALB/c mice with the recombinant plasmid for Hantaan virus can induce not only the systemic specific immune responses,but also high mucosal immune responses.It suggests intranasal immunization is a better route of inoculation with a potential application value.
