家兔混合反流性食管炎动物模型的制作

摘    要:目的通过酸和胆汁混合灌注制备家兔混合反流性食管炎动物模型。方法采用盐酸和猪胆汁混合灌注建立家兔混合反流性食管炎模型,模型组以灌注不同时间分为4 d组、7 d组及10 d组,正常组则灌注生理盐水;分别在肉眼下、光镜及电镜下观察各组食管变化,并进行分级比较。结果混合灌注后食管下端病理改变明显。3组模型家兔食管下段存在不同程度黏膜粗糙、充血、黏膜下血管增生,见散在或广泛糜烂,并可见不同程度溃疡。3组模型家兔食管肉眼积分差异有统计学意义(P〈0.01)。模型家兔可见部分鳞状上皮固缩脱落,固有层嗜酸性细胞、中性粒细胞浸润,黏膜下层内血管扩张、充血,见不同程度的溃疡、组织坏死,食管肌层也可见不同程度炎性细胞浸润。3组模型家兔食管病理积分差异有统计学意义(P〈0.05)。电镜下模型家兔见不同程度线粒体肿胀、肌质网扩张。结论酸和胆汁混合灌注可以成功制备家兔混合反流性食管炎动物模型。

Abstract:Objective To establish the rabbit model of mixed reflux esophagitis by mixed perfusion of hydrochloric acid and bile. Methods The rabbit model of mixed reflux esophagitis was established by mixed perfusion of hydrochloric acid and bile. The model groups were divided into four-day, seven-day,ten-day group.The normal group was perfused with the solution of Sodium Chloride. The esophagus pathological changes in the different groups were observed by macroscopic observation, the optical microscope, and the electronic microscope. And the pathological grade of the different groups were compared. Results Significant pathological changes were observed in the lower esophageal bottom. The general form by macroscopic observation: The mucosa in the lower part of the esophagus in the normal group was smooth, and there was no blood vessel hyperplasia under the mucosa. The esophageal mucosa in the model group existed diverse levels of rough and hyperemia, and blood vessel hyperplasia under the mucosa, sporadic or extensive erosion and diverse levels of ulcer were observed too. The score of macroscopic observation among three model groups were significantly different (P〈0.01). under the optical microscope,there was no pathological changes in the mucosa and the muscle layers in the normal group. In the model group, there were diverse levels of pyknosis and defluxion of squamous epithelium, the infiltration of eosinophile and neutrophil, dilation and hyperemia in the submucosa, and diverse levels of ulcer and tissue necrosis, as well as diverse levels of inflammatory cell infiltration of the muscle layer was observed. The pathological scores in the three model groups were significantly different (P〈0.05). Under the electronic microscope, there were diverse levels swelling of the chondriome and dilation of the peripheral sarcoplasmic reticulum. Conclusion It is successful to establish the rabbit model of the acid and bile mixed reflux esophagitis by mixed perfusion.
