赖诺普利对糖尿病模型大鼠肾脏的保护作用

摘    要:目的探讨血管紧张素转换酶抑制剂赖诺普利对糖尿病大鼠肾脏病变的保护作用。方法24只健康雄性Wistar大鼠随机分成正常对照组(NC组,n=8)、糖尿病未治疗组(DM组,n=8)、糖尿病苯那普利治疗组(DL组,n=8)。应用链脲佐菌素(STZ)诱导糖尿病模型,造模成功后,用赖诺普利(10mg·kg^-1·d^-1)对DL组进行治疗性灌胃,其余2组均用等量自来水灌胃。10周治疗后,观察了其对糖尿病大鼠肾脏的保护作用。结果对照组糖尿病大鼠肾小球滤过率(GFR)、肾血流量(RPF)、滤过分数(FF)及尿白蛋白(Upro)均较正常对照组明显升高,赖诺普利治疗后糖尿病大鼠GFR、RPF、FF及UA均较糖尿病对照组明显降低,血糖、糖化血红蛋白较糖尿病对照组无显著性差异。结论早期应用赖诺普利能改善糖尿病大鼠肾脏的血流动力学异常,减少尿蛋白排泄,防止或延缓糖尿病肾脏的病理学进展。

Abstract:Objective : rio investigate the protective effect of Lisinopril on diabetic nephropathy (DN) in rats. Methods : Twenty-four healthy male Wistar rats were randomly divided into the normal control group( NC group), the diabetic group untreated with Lisinopril (DM group) and the diabetic group treated with Lisinopril (DL group). The DN model was established following intraperitoneal injection of streptozocin (60 mg·kg^-1 ). Lisinopril ( 10mg·kg^-1·d^-1 ) was given to the DL group for 10 weeks, whereas the same milliliter water was given to the other two groups,and the protective effects of Lisinopril on renal function were evaluated. Results: The glomerular filtration rate( GFR), renal plasma flow(RPF) ,filtration fraction (FF) and urinary protein excretion significantly increased in diabetie rats. After Lisinopril tratement, the GFR, RPF, FF and urinary protein(Upro) were markedly reduced as compared with those untreated diabetic rats. Conclusion: Early treatment with Lisinopril can normalize the abnormal renal hemodyamics, reduce the urinary protein excretion and prevent renal parhological progress in diabetic rats.
