摘    要:目的 探讨银离子(Ag+)、铜离子(Cu2+)、锌离子(Zn2+)对5种常见念珠菌(白色念珠菌、热带念珠菌、近平滑念珠菌、克柔念珠菌和光滑念珠菌)的抗菌作用。方法 使用去离子水分别将硝酸银、氯化铜和氯化锌配制为0.1 mol·L-1的水溶液。取保存的白色念珠菌标准菌株ATCC-60193、近平滑念珠菌、光滑念珠菌、热带念珠菌、克柔念珠菌,加入溶菌肉汤培养液,使用比浊仪调配菌液浊度为0.5麦氏单位,然后使用无菌去离子水将菌液稀释至106 CFU·L-1。取配制好的0.1 mol·L-1的硝酸银、氯化铜和氯化锌溶液,分别倍比稀释为6个浓度(10-2、10-3、10-4、10-5、10-6、10-7 mol·L-1),分别取各浓度的金属离子溶液100μL与预制好的5种念珠菌悬液100μL进...

Abstract:Objective To explore the antibacterial effect of silver ions (Ag+),copper ions (Cu2+) and zinc ions (Zn2+) on five common candida species (Candida albicans,Candida tropicalis,Candida parapsilosis,Candida krusei and Candida glabrata).Methods Silver nitrate,copper chloride and zinc chloride were prepared into 0.1 mol·L-1 aqueous solution with deionized water.The preserved standard bacteria beads of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis,Candida krusei were taken and added into the culture solution of bacteriolytic broth.The turbidity of bacterial solution was prepared to 0.5 MCU by using the turbidimeter,and then the bacterial solution was diluted to 106 CFU·L-1 by using sterile deionized water.The prepared 0.1 mol·L-1 silver nitrate,copper chloride and zinc chloride solutions were taken and diluted to 6 concentrations( 10-2,10-3,10-4,10-5,10-6 ,10-7 mol·L-1) in multiple proportions,respectively.The 100 μL metal ion solutions of each concentration and 100 μL five kinds of prefabricated candida suspension were mixed at 1∶1 (volume ratio),and then placed in 96 well plates,and 6 sample groups were seted;at the same time,the 0 mol·L-1 blank group was set(100 μL sterilized deionized water was taken and placed in 96 well blank plate,and 100 μL candida suspension was added for mixing,then was cultured at 37 ℃ for 48 hours).The co-culture solution was blowed,sucked and mixed,then was diluted to 100 times and 1 000 times with sterile deionized water.The 100 μL solution was taken and smeared on Sabouraud′s fungus culture medium for culturing of 48 hours at 37 ℃,then the number of bacterial colony on the plate was recorded and the sterilization rate was calculated.If the sterilization rate ≥ 99% indicated strong antibacterial effect,and if the sterilization rate ≥ 90% indicated it had antibacterial effect.Results When the Ag+ concentration ≥ 10-5 mol·L-1,there was no bacterial colony growth after Ag+ acted the standard beads of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis and Candida krusei,and the sterilization rates were (100.00 ± 0.00)%.There was no bacterial colony growth of Candida krusei and Candida glabrata when Ag+ concentration was 10-6 mol·L-1,and the sterilization rates were (100.00 ± 0.00)%.There were some bacterial colony growth of the standard beads of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis and Candida krusei after action of 10-7 mol·L-1 Ag+,and the sterilization rates were less than 90%.When the Cu2+ concentration ≥10-4 mol·L-1,there was no bacterial colony growth after Cu2+ acted the standard beads of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis and Candida krusei,and the sterilization rates were (100.00 ± 0.00)%.There was no bacterial colony growth of the Candida tropicalis,Candida parapsilosis and Candida glabrata after action of 10-5 mol·L-1 Cu2+,the sterilization rates were (100.00 ± 0.00)%.When Cu2+ concentration was 10-6 mol·L-1,the sterilization rate of Candida parapsilosis was more than 90%.There were bacterial colony growth of the standard beads of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis and Candida krusei at different levels after action of 10-7 mol·L-1 Cu2+,and the sterilization rates were less than 90%.When the Zn2+ concentration ≥ 10-3 mol·L-1,there was no bacterial colony growth after Ag+ acted the standard beads of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis and Candida krusei,and the sterilization rates were (100.00 ± 0.00)%.When the Zn2+ concentration was 10-4 mol·L-1,there was no bacterial colony growth of the standard beads of Candida albicans ATCC-60193,Candida tropicalis,Candida parapsilosis and Candida krusei,and the sterilization rates were (100.00 ± 0.00)%;the sterilization rate of Zn2+ on Candida glabrata was (99.25 ± 0.74)%.When the Zn2+ concentration ≤10-5 mol·L-1,there were bacterial colony growth of the standard beads of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis and Candida krusei at different levels after the action of Zn2+,and the sterilization rates were less than 90%.Conclusion Ag+,Cu2+ and Zn2+ have certain inhibitory effects on the growth of five common candida species of Candida albicans ATCC-60193,Candida parapsilosis,Candida glabrata,Candida tropicalis and Candida krusei,but the sterilization rate decreases with the decrease of ion concentration,and the optimal antibacterial concentration of the same metal ion to different candida species is different.The lowest concentrations of Ag+,Cu2+ and Zn2+ with simultaneously antibacterial effects on five common candida species are 10-5 ,10-4,10-4 mol·L-1,respectively;among them,Ag+ has stronger and broader antibacterial properties.
