当归加玻璃酸钠关节腔注射对兔膝骨关节炎软骨细胞Bcl-2、Bax表达的影响

目的:研究当归注射液结合玻璃酸钠注射液关节腔注射对兔膝骨关节炎软骨软骨细胞Bcl-2、Bax表达的影响。方法:健康家兔50只随机分为5组,正常对照组、模型对照组、当归组、玻璃酸钠组、当归与玻璃酸钠结合组,每组10只,后4组按Hulth方法建立兔膝骨性关节炎的动物模型,术后每周经膝关节穿刺给药1次,连续给药6周后处死、取材,关节软骨作大体及光镜观察,检测软骨细胞Bcl-2(B-cell lymphomas,B淋巴细胞-2蛋白)、Bax(Bcl-2 Associated X protein,B淋巴细胞-2的相关X蛋白)的表达灰度。结果:各药物预防组动物关节软骨的大体观察评分、组织学评分、软骨细胞Bax的表达灰度均低于模型组(均P<0.01),软骨细胞Bcl-2的表达灰度均高于对照组(均P<0.01),当归与玻璃酸钠合用组效果更佳(均P<0.05)。结论:当归注射液和玻璃酸钠注射液关节腔注射对软骨细胞的退变有显著的减缓作用,尤以两者合用时显著。

Effect of Angelica and Sodium Hyaluronate(SH)by Intra-articular Injection on the Degenerative Cartilage of Osteoarthritis to the Expression of Bcl-2, Bax

Objective:To study the effect of Angelica and Sodium Hyaluronate(SH)to the expression of Bcl-2,Bax of zhe cartilage of osteoarthritis in rabbits.Methods:A total of 50 adult rabbits were randomly divided into five groups-normal control group,model control group,Angelica treatment group,SH treatment group,AG and SH treatment group.Animal models of osteoarthritis on the knees of rabbits in the late four groups were established by Hulth.Corresponding medicines were injected into the knee joint cavity respectively once every a week after 1 week of the operation.All the animals were killed 6 weeks later and samples of the cartilage,plasma and synovium were taken.Macroscopical and histological examinations of the cartilage were performed and immunohistochemical analyses for Bcl-2 and Bax in the cartilage were done.Result:The macroscopical and histological grade of the cartilage,the gray scores of Bax in the cartilage cells in all prophylactic groups were lower than ones in model groups(P<0.01 respectively).The gray scores of Bcl-2 in the cartilage cells were higher than ones of the control group(P<0.01 respectively).But the therapeutic effect in groups by being injected both Angelica and sodium hyaluronate was optimal in three groups(P<0.05 respectively).Conclusion:The intra-articular injection by Angelica and sodium hyaluronate may reduce the degeneration of the cartilage,and the effect is very remarkable.
