摘    要:目的探究miR-28-5p对结肠癌细胞增殖和迁移能力的影响。方法采用qRT-PCR检测miR-28-5p在正常结肠细胞NCM460与结肠癌细胞HCT116、SW480、SW620中表达量的差异。将HCT116、SW480、SW620各分为三组:对照组,过表达组,沉默组。对照组不予特殊处理,其余两组分别转染miR-28-5p mimic和miR-28-5p inhibitor,采用qRT-PCR验证转染效率,采用CCK8法检测三组细胞的增殖能力,采用划痕实验检测三组细胞的的迁移能力。结果miR-28-5p在结肠癌细胞HCT116、SW480、SW620中低表达,与正常结肠细胞NCM460比较差异有统计学意义(P<0.05)。与对照组相比,过表达组细胞增殖和迁移能力均降低(P<0.05),沉默组细胞增殖和迁移能力均增强(P<0.05)。结论miR-28-5p在结肠癌细胞中低表达且有调控结肠癌细胞增殖和迁移的作用。

Abstract:【Objective】To investigate the effect of miRNA-28-5p on the proliferation and migration of colon cancer cells.【Methods】qRT-PCR was used to detect the expression of miR-28-5p in normal colon cells NCM460 and colon cancer cells line HCT116,SW480,SW620,HCT116,SW480 and SW620 were divided into control group,mimics group and inhibitors group.The control group was not treated,and the remaining two groups were transfected with miR-28-5p mimic and miR-28-5p inhibitor respectively,qRT-PCR was used to detect the transfection efficiency.CCK8 method was used to detect the proliferation ability of the three groups of cells,and scratch test was used to detect the migration ability of the three groups of cells.【Results】The expression of miR-28-5p in HCT116,SW480 and SW620 cells was significantly lower than that in normal colon cells NCM460(all P<0.05).In contrast to the control group,the proliferation and migration ability of cells in mimics group was decreased(all P<0.05),and the proliferation and migration ability of cells in inhibitors group was enhanced(all P<0.05).【Conclusion】miR-28-5p has low expression in colon cancer cells and can regulate the proliferation and migration of colon cancer cells.
