Abstract:Objective It is to observe the effect of Lycium barbarum polysaccharide on the expression of insulin-like growth factor-2(IGF-2) and insulin-like growth factor-binding protein-2(IGFBP-2) in blood and liver tissue of hepatic fibrosis rats, and to explore the inhibition of hepatic fibrosis by Lycium barbarum polysaccharide. mechanism of action. Methods Eighteen healthy SD rats were selected, and the liver fibrosis rat model was prepared by common bile duct ligation. The successful model rats were randomly divided into the model group and the Lycium barbarum polysaccharide group, 6 rats in each group, and 6 normal SD rats of the same species were selected as a control group. The Lycium barbarum polysaccharide group was given intragastric administration of Lycium barbarum polysaccharide 30 μg/g every day, and the model group and the control group were given the same amount of normal saline by intragastric administration, all treated for 30 days. The structure of liver tissue in each group was observed by HE staining, the levels of IGF-2 and IGFBP-2 in serum were detected by enzyme-linked immunosorbent assay, and the protein expressions of IGF-2 and IGFBP-2 in liver tissue were detected by Western blotting. Results Compared with the control group, necrotic areas of different sizes, and connective tissue hyperplasia in the portal area were found in the liver tissue of the rats in the model group;the levels of serum IGF-2 and IGFBP-2 and the protein expressions of IGF-2 and IGFBP-2 in the liver tissue were significantly increased(P<0.05). Compared with the model group, the necrotic area in the liver tissue of the rats in the Lycium barbarum polysaccharide group and the components of the connective tissue in the portal area were decreased;the levels of serum IGF-2 and IGFBP-2 and the protein expressions of IGF-2 and IGFBP-2 in the liver tissue were significantly decreased(all P<0.05). Conclusion Lycium barbarum polysaccharide can alleviate liver fibrosis injury by regulating the expression of IGF-2 and IGFBP-2 in blood and liver tissue.
