卡介菌多糖核酸膀胱灌注对膀胱癌细胞端粒酶活性及细胞凋亡的影响

摘    要:目的:探讨卡介菌多糖核酸(BCG—PSN)膀胱灌注对膀胱癌细胞端粒酶活性及细胞凋亡的影响。方法132例膀胱移行细胞癌患者术前膀胱灌注BCG—PSN20mg,隔天1次,共3次。对术前术后的标本采用端粒重复序列扩增法(TRAP—PCR—EI.ISA)和末端脱氧核苷酸转移酶介导缺口末端标记法(TUNEI。)分别检测肿瘤细胞端粒酶活性及细胞凋亡的情况。结果:膀胱灌注前后肿瘤细胞端粒酶活性分别为(12.78±3.67)和(3.89±1.12),差异有统计学意义(P〈0.05)。灌注前后肿瘤细胞凋亡指数分别为(31.6±11.3)%和(69.1±12.4)%,差异有统计学意义(P〈0.05)。结论:BCG—PSN膀胱灌注能抑制膀胱癌细胞端粒酶活性,并诱导肿瘤细胞凋亡,具有明显的抗肿瘤作用。

Abstract:Objective:To study the telomerase activity and cellular apoptosis in patients with bladder cancer before and after intravesical instillation with polysacharide nucleic acid fraction of BCG(BCG-PSN).Methods:The telomerase activity and cellular apoptosis of 32 cases of bladder cancer tissues were detected by terminal deoxynucleotidyl-transferase mediated dUTP-biotin nick end labeling(TUNED and telomeric repeat amplification protocol(TRAP-PCR-ELISA) assay both before and after intravesical instillation with BCG-PSN.Results:The telomerase activity for bladder cancers before and after treatment with BCG-PSN was(12.78±3.67) and(3.89±1.12) respectively(P<0.05).The apoptosis index for bladder cancers before and after treatment with BCG-PSN was(31.6±11.3)%and(69.1±12.4)%respectively(P<0.05).Conclusions:BCG-PSN is effective in inhibiting the telomerase activity and in inducing cellular apoptosis for bladder cancer.It obviously plays an important role in anti-cancer.
