蚕梅方提取物对AOM/DSS诱发腺瘤性息肉小鼠的防治作用

摘    要:
目的:探讨蚕梅方(由僵蚕、乌梅组成)提取物对AOM/DSS诱发小鼠腺瘤性肠息肉的防治作用。方法:以C57BL/6小鼠为研究对象,随机分为正常组、模型组、阳性对照组(阿司匹林组)、蚕梅低剂量组(0.65 g/ml)、蚕梅高剂量组(2.6 g/ml)。除正常组外,其余各种采用AOM/DSS诱发小鼠结直肠腺瘤性息肉模型,并于造模过程中各药物组予相应的药物灌胃干预。实验结束,比较各组结直肠腺瘤个数及抑瘤率,并以免疫组化法检测结肠黏膜细胞周期蛋白D1(Cyclin D1)表达水平;应用ELISA检测小鼠血浆白介素-6(IL-6)表达水平。结果:经93 d灌胃给药,阳性对照组、蚕梅高剂量组、蚕梅低剂量组腺瘤抑制率分别为14.49%、70.45%、8.78%,与阳性对照组比较蚕梅高剂量组有显著抑制腺瘤作用(P0.05),且蚕梅高剂量组能显著降低Cyclin D1阳性发生率(P0.05);与正常组比较,模型组和阳性对照组IL-6高表达,蚕梅方低剂量组和高剂量组可显著下调IL-6表达(P0.05)。结论:蚕梅方提取物能显著降低AOM/DSS小鼠腺瘤性息肉的发生率,且有明显的量效关系;其作用机制可能与抑制Cyclin D1、显著下调IL-6表达有关。
