| 摘 要： | 目的 研究他克莫司在中国肾移植患者的体内药动学。方法 12例肾移植患者口服他克莫司达稳态后,用ELISA法测定全血中他克莫司浓度。用3P97程序拟合计算药动学参数。结果 口服他克莫司的药-时曲线符合一级吸收一室模型.全血浓度达峰时间为(1.54±0.58)h,峰浓度为(32.98±26.58)μg·L-1,消除t1/2为(13.3±3.9)h,AUC0～12为(154.60±89.13)μg·h·L-1。12 h谷浓度与AUC0～12有较好的相关性。结论 他克莫司个体间药动学差异大,与国内外文献报道基本一致。

| Abstract: | OBJECTIVE To study the pharmacokinetics of tacrolimus in renal transplant recipients. METHODS After oral administration of tacrolimus, the blood concentrations of tacrolimus were measured by ELISA. The pharmacokinetic parameters of tacrolimus were estimated by 3P97 program. RESULTS The blood concentration-time profiles conformed to one compartment model with first order absorption.The tmax was( 1.54±0.58) h, cmax(32.98±26.58) μg·L-1,t1/2β(13.3±3.9)h and AUC0～12( 154.60±89.13) μg·h·L-1, respectively. The trough concentrations at twelve hours were correlated well with AUC0～12. CONCLUSION The pharmacokinetics of tacrolimus was highly variable between individuals in renal transplant recipients, which was concordant with that of the literature reports. |
