摘    要:
目的:探讨转化生长因子β3(TGFβ3)mRNA在小鼠磨牙牙胚发育过程中的表达及其意义。方法:制备小鼠磨牙各发育阶段标本,用原位杂交分析TGFβ3mRNA在牙胚中的表达与分布。结果:TGFβ3mRNA在小鼠磨牙牙胚不同发育阶段呈时间-空间特异性模式表达。TGFβ3mRNA在蕾状期和帽状期的上皮强表达,在牙胚发育晚期的成牙本质细胞和成釉细胞的表达随细胞分化程度而增强。结论:TGFβ3mRNA在牙形态发生过程中可能通过自分泌和旁分泌机制协同调控上皮-间充质相互作用及成牙本质细胞和成釉细胞的分化。

Abstract:
Objective: To investigate the temporal and spatial expression and localization of transforming growth factor-β3 mRNA during mouse molar development.Methods: The distribution pattern of TGFβ3 mRNA was analyzed on 5 μm serial sections of paraformaldehyde-fixed and paraffin-embedded embryonic mouse heads or mandibles using in situ hybridization.Results: The results showed that TGFβ3 mRNA was expressed during mouse molar development in a temporally and spatially regulated fashion.TGFβ3 mRNA was strongly expressed in dental epithelia at bud and cap stage.The expression of TGFβ3 mRNA was increased in the layer of odontoblast and ameloblast according with the differentiation of these cells.Conclusion: These results suggest that TGFβ3 mRNA may have an important role in tooth development and be involved in an autocrine and paracrine fashion in the epithelial-mesenchymal interactions regulating morphogenesis.
