酸性染料比色法测定飞龙掌血提取物中总生物碱的含量

目的:建立飞龙掌血提取物中总生物碱含量测定的方法。方法:通过酸性染料比色法,利用紫外分光光度计进行飞龙掌血总生物碱的含量测定,并进行方法学考察,建立适宜的含量测定方法。结果:建立了飞龙掌血总生物碱含量测定方法;于330 nm的检测波长下,以氯化两面针碱(Nitidine Chloride)为对照品,在1.497 6～5.990 4μg/ml范围内对照品含量与吸光度呈良好的线性关系;回归方程为A=0.115 9c+0.137 6(r=0.999 7);日内精密度RSD=0.53%、日间精密度RSD=2.57%;加样回收率为100.08%,RSD=3.30%。结论:该方法回收率、稳定性、重现性、精密度良好,可用于飞龙掌血总生物碱的含量测定。

Determination of Total Alkaloids in Radix Toddaliae Asiaticae by Acid Dye Colorimetry

Objective: To establish a method for quantitative determination of the total alkaloids in Radix Toddaliae Asiaticae.Methods: Acid dye colorimetry and ultraviolet spectrophotometer were used to detect the content of total alkaloid in Radix Toddaliae Asiaticae with methodology research.The proper method for the determination was established.Results: The method for total alkaloid content determination was established;the result showed a good linear relationship between the absorbance and quantity of Nitidine Chloride to the total alkaloids by acid dye colorimetry at 330 nm wavelength.Within the range of(1.497 6~5.990 4)μg/ml,the regression equation was A=0.115 9c+0.137 6(r=0.999 7);the RSD of within-day precision was 0.53%,day to day precision was 2.57%;average recovery was 100.08%,RSD was 3.30%.Conclusion: This method is accurate,reliable and simple,which can be used for content determination of total alkaloids in Radix Toddaliae Asiaticae.
