胸腺肽对慢性阻塞性肺病患者树突状细胞的免疫调节

摘    要:
目的:探讨胸腺肽对慢性阻塞性肺病(COPD)患者树突状细胞的免疫调节作用.方法:分离COPD患者和健康者外周血单个核细胞并诱导成树突状细胞,在体外通过胸腺肽干预树突状细胞分为四组,分别为COPD胸腺肽刺激组、COPD组、健康者胸腺肽刺激组、健康组,通过EHSA方法检测各组外周血树突状细胞分泌Th1型细胞因子IFN-γ和Th2型细胞因子IL-4的表达水平.结果:COPD胸腺肽刺激组较COPD组和健康者胸腺肽刺激组IFN-γ明显升高(分别P<0.05和P<0.01),而COPD胸腺肽刺激组较COPD组IL-4的表达明显降低(P<0.05),COPD组较健康组分泌细胞因子也明显增多(P<0.01).结论:胸腺肽可调节COPD患者IFN-γ、IL-4的分泌表达,有利于改善COPD患者的免疫功能.

Abstract:
Objective: To investigate immune regulation of Thymosin on dendritic cells from patients with COPD. Methods:We isolated peripheral blood mononuclear cells of patients with COPD and healthy donors and transferred them into dendritic cells. These ceils were divided into four groups, which contained thymosin-stimulated dendritic cells and non-thymosin-stimulated dendritic cells respectively from patients with COPD and healthy donors. The levels of Thl cytokines IFN-'y and Th2 cytokines IL-4 were examined from peripheral blood dendritic cells of four groups using ELISA.Results:The IFN-γ level from thymosin-stimulated dendritic ceils of patients with COPD was increased compared to non-thymosin-stimulated dendritic cells of patients with COPD and thymosin-stimulated dendritic cells of healthy donors (respectively P〈0.05 and P〈0.01),but the IL-4 level from thymosin-stimulated dendritic cells of patients with COPD was decreased compared to non-thymosin-stimulated dendritic cells of patients with COPD (P〈 0.05). The level of cytokines from non-thymosin-stimulated dendritic cells of patients with COPD was increased compared to healthy donors. Conclusion: This study indicates that thymosin can regulate the levels of IFN-γ and IL-4 from patients with COPD in vivo, it is beneficial to improve the immune function of patients with COPD.
