Toll样受体2 Arg677Trp和Arg753Gln基因多态性与梅毒治疗后血清学试验阳性相关性研究

 目的:研究TLR2胞内信号传导区域内Arg677Trp、Arg753Gln基因多态性是否与梅毒治疗后患者的血清学阳性结果相关。方法:采用PCR RFLP对实验组1的100例梅毒血清固定患者、实验组2的50例梅毒治疗后TPPA阳性而TRUST阴性的患者及100例健康对照者进行检测；部分判定困难的标本进一步采用DNA测序证实。结果:所有样本均未检测到TLR2 Arg677Trp基因多态性。对照组无Arg753Gln基因多态性，实验组1有1例（1%）TLR2 Arg753Gln基因多态性，与对照组比较无明显差异（ X²=1.01，P=0.316）；实验组2有6例（12%）TLR2 Arg753Gln基因多态性，与对照组比较有明显差异（ X²=12.50，P＜0.01）。结论:目前未发现TLR2 Arg677Trp和Arg753Gln基因多态性与梅毒血清固定相关。梅毒规范治疗后出现TPPA阳性TRUST阴转的现象，可能与TLR2Arg753Gln基因多态性相关。

Correlation of TLR2 Arg677Trp and Arg753Gln polymorphisms with post treatment serological test positive in syphilis patients

Objective：To investigate whether the Arg677Trp and Arg753Gln polymorphisms in the intracellular signaling region of TLR2 are linked to post treatment serological test positive in syphilis patients. Methods:PCR RFLP was used to detect TLR2 Arg677Trp and Arg753Gln polymorphisms in 100 syphilis patients with sero fixation (group 1), 50 patients with TPPA positive while TRUST negative after syphilis treatment (group 2), and 100 healthy controls. DNA sequencing was used to confirm the diagnosis in case of diagnostic uncertainty. Results:TLR2 Arg677Trp polymorphism was not detected in any of the samples. While only one patient in group 1(1%) displayed TLR2 Arg753Gln polymorphism, proportion of patients with TLR2 Arg753Gln polymorphism was significantly higher in group 2 than in the controls (6/50 vs. 0,   X²=12.50，P＜0.01). Conclusions: Neither TLR2 Arg677Trp nor Arg753Gln polymorphisms is associated to syphilis serum fixation. However, TLR2 Arg753Gln polymorphism could be linked to post treatment TPPA positive and TRUST negative in syphilis patients.
