定量测定45例血小板膜糖蛋白及其在急性脑梗死中的意义

目的:探讨流式细胞术定量测定血小板膜糖蛋白在急性脑梗死中的临床意义。方法:应用流式细胞仪测定静息状态和TRAP激活的血小板所结合单抗的分子数。结果:急性脑梗死患者血小板静息状态下GPⅢa和GPⅡb/Ⅲa密度分别为(76.8±14.1)×103/血小板、(73.9±10.5)×103/血小板]高于健康对照组分别为(55.6±5.8)×103/血小板、(50.5±4.1)×103/血小板,P<0.05],TRAP活化后患者血小板GPⅢa和GPⅡb/Ⅲa密度分别为(102.6±16.4)×103/血小板、(108.5±16.7)×103/血小板]亦明显高于对照组分别为(69.7±18.3)×103/血小板、(61.2±5.7)×103/血小板,P<0.01];患者血小板静息状态下,GMP-140表达与对照组比较没有显著性差异(P>0.05),而TRAP激活后患者血小板GMP-140表达(1.21±0.41)×103/血小板]明显高于对照组(0.87±0.26)×103/血小板,P<0.05];患者血小板静息和活化状态GPⅠb表达与对照组比较没有显著性差异(P>0.05)。结论:急性脑梗死患者血小板呈较为活化状态,血小板膜糖蛋白测定对病情的分析和疗效的判断有重要的临床参考价值。

Quantitative analysis of platelet membrane glycoprotein and its clinical significance in acute cerebral infarction

Objective:To explore the flow cytometric quantitative analysis of platelet membrane glycoprotein and its clinical significance in acute cerebral infarction.Methods:The molecule numbers of platelet GPⅠb,GPⅢa,GPⅡb/Ⅲa and GMP-140 on resting platelets and platelets activated by TRAP were detected by Flow cytometry.Results:Whether on resting platelets or platelets activated by TRAP,the surface densitits of GPⅢa and GPⅡb/Ⅲa in patients with cute cerebral infarction were significantly higher than those in healthy individuals(P<0.05,P<0.01).But there was no significant difference between patients and healthy individuals on GMP-140 expression of resting platelets(P>0.05).On activated platelets the density of GMP-140 in patients was higher than that in healthy individuals(P<0.05).There was no statistic differvence in the surface density of GPⅠb between patients and control(P>0.05).Conclusions:The platelets in patients with cute cerebral infarction are on highly activated status.Quantitative detection of platelet membrane glycoprotein by Flow cytometry is of highly clinical value.
