草履虫的扫描电镜观察

摘    要:本文描述用二甲亚砜(DMSO)冷冻割断法和改进的样品制备技术制备草履虫标本后,扫描电镜下的草履虫虫体形状、纤毛、表膜、纤维、刺丝泡、伸缩泡、大核、口沟、胞咽、食物泡、各种颗粒等三维的微细结构以及生殖与体形变化的表面结构。

Abstract:With the method of dimethyl sulfoxide(DMSO)freeze-fracture and technique of specimen preparation and observation with the scanning electron microscope (SEM), we obtained directly morphological information about the three-dimensional fine structures, such as cilia, surface membrane, fibrae, trichocyst, contractile vacuole, food vacuole, various particles, macronucleus, oral groove, cytopharynx, etc. In paramecia some of these fine Structures can not be seen with the optic microscope or the transmission electron microscope. Thus, some new informations can be shown by SEM.
