双价志贺疫苗滴鼻免疫小鼠后粘膜免疫与系统免疫应答持续时间的观察

摘    要:本文探讨双价志贺疫苗滴鼻免疫小鼠一段时间后,粘膜免疫和系统免疫应答的变化。将BALB/c小鼠随机分为三组,每组30只。PBS、FSM-2117和FS-5416(菌量为5×10~6、1×10~7、4×10~7和4×10~7CFU/只/次)经滴鼻途径免疫小鼠。间隔2周,4次免疫后7、30和90d活杀,收集鼻咽、肺、肠、生殖道冲洗液和血清。采用ELISA法检测其中特异性抗福氏、宋内LPSIgA和IgG。结果是两株疫苗经鼻内免疫后,诱发鼻咽、肺、胃肠道和生殖道等不同粘膜部位及血清中特异性抗福氏、宋内LPSIgA、IgG的显著增加(P<0.01)。特异性抗体水平虽然在免疫后30、90d明显下降,但仍明显高于PBS对照组水平。故认为两株双价志贺疫苗滴鼻免疫小鼠后能有效诱导粘膜免疫和系统免疫应答,并持续较长时间。

Abstract:To observe the fluctuation of mucosal and systemic immune responses after intranasal immunization of mice with bivalent Shigella vaccines, BALB/c mice were intranasally immunized with PBS, FSM-2117 or FS-5416 (5 × 106, 1×107, 4×107, 4× 107CFU/mouse/dose )in 4 doses at an internal of two weeks. The nasopharyngeal, lung, in-testinal, genital tract lavage fluids and sera were taken on the 7l , 30' and 90' day since the last immunization, and the specific IgA or IgG against F2a or Sonne-LPS in these specimens were assayed by ELISA. It was found that the specific IgA and IgG responses of mice against the LPS of Sh .flexner and Sh. sonne in the different mucosal sites and in the sera increased significantly on the 7th day after intranasal immunization with these two Shigella vaccines in comparison with those of the control mice. Although this immune response decreased markedly on the 30 and 90 day after immunization , but it was stili higher than those of the control mice. it concludes that the intranasal immunization of mice with two bivalent Shigella vaccines can induce effectively the mucosal and systemic immune responses and these responses can persist for a comparatively long time.
