福建汉族人群STR位点遗传多态性分析及其在亲子鉴定中的应用

摘    要:目的调查福建汉族人群的D5S818、FGA、D3S1358、THOI、D13S317、D16S539、D2S1338、D8S1179、D21S11、D7S820、CSF1PO、D19S433、vWA、TPOX、D18S51共15个STR位点多态性,探讨STR基因分析在亲子鉴定中的应用价值。方法应用PCR复合扩增和五色荧光Genescan技术对210个福建汉族个体和89例亲子鉴定案件的15个STR位点作基因分型。结果福建汉族人群15个STR位点的等位基因频率、基因型分布符合Hardy-Weinberg平衡。D5S818、FGA、D13S317、D16S539、D2S1338、D8S1179、D21S11、D7S820、D19S433、vWA、D18S51位点属于高鉴别力、高杂合度、高信息含量的STR位点。15个STR位点的累积个体识别能力(DP)、非父排除率(PE)均为0.9999以上。89例亲子鉴定案例中,否定亲权的25个案例中均至少有3个位点不符;认定亲权的64例子中,亲子关系指数(RCP)大于99.73%。结论获得了福建汉族人群中15个STR位点的遗传多态性数据。应用这15个STR位点的等位基因分析,能成功进行亲子鉴定,显著提高亲子关系指数。

Abstract:Objective To study the genetic polymorphism of 15 STR loci including D5S818,FGA,D3S1358,THOI,D13S317,D16S539,D2S1338,D8S1179,D21S11,D7S820,CSF1PO,D19S433,vWA,TPOX and D18S51 among Han population in Fujian and their application in parentage testing.Methods The 15 STR loci of 210 unrelated individuals of Han population in Fujian were amplified by multiple PCR and analyzed by Genescan software.Results All allele frequencies were calculated and their distributions were in accordance with Hardy-Weinberg equilibrium.The Index of combined discrimination power,polymorphism information content,heterozygosity and power of exclusion were greater than 0.9999.The paternity of 25 in 89 cases were excluded by parentage testing,and more than three incoordinate loci were found in each cases.Among the 64 cases of determined paternal relation,the relative chance of paternity (RCP) was greater than 99.73%.Conclusion We obtained all the allele frequencies of these 15 STR loci among Han population in Fujian.Our result suggest that STR genetic study can be applied to parentae testing,and it will increase the RCP obviously.
