新型冠状病毒灭活方式对高效液相色谱法监测万古霉素血药浓度的影响

摘    要:
目的考察新型冠状病毒不同灭活方式对高效液相色谱(HPLC)法监测万古霉素血药浓度的影响。方法经56℃水浴加热0 min和30 min,以及紫外照射0,30,60 min后,采用HPLC法测定质控液和患者血样中万古霉素的质量浓度。结果56℃水浴加热30 min不会影响监测结果;紫外照射30 min和60 min后,质控液和患者血样中万古霉素均降解,对监测结果影响较大。结论采用HPLC法监测新型冠状病毒肺炎(COVID-19)患者万古霉素血药浓度时,可用湿热法56℃水浴30 min灭活病毒,不可用紫外照射灭活病毒,且万古霉素质控液与患者血样均要注意避免紫外照射。

Abstract:
Objective To investigate the effect of different inactivation methods of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)on monitoring vancomycin blood concentration by high performance liquid chromatography(HPLC).Methods The HPLC method was used to determine the concentration of the quality control solution and the vancomycin blood concentration of the patients after heating at 56℃for 0,30 min and 0,30,60 min of UV irradiation.Results Heating in 56℃water bath for 30 min did not affect the monitoring results of vancomycin blood concentration in patients.After 30,60 min of UV irradiation,vancomycin quality control solution and blood samples of patients were degraded,which greatly affected the monitoring results.Conclusion To monitor the concentration of vancomycin in the blood of patients with coronavirus disease 2019(COVID-19)by HPLC,the virus can be inactivated by soaking in 56℃water for 30 min,but not by UV irradiation.In addition,the vancomycin quality control solution and blood samples should avoid UV radiation.
