Dkk3在肝细胞肝癌中的表达及其临床意义

目的探讨dkk3在肝细胞肝癌中的表达及临床意义。方法 40例肝癌及其癌旁组织,10例正常肝组织采用Real time PCR检测DKK3mNA水平表达情况和228例肝癌组织及癌旁组织石蜡切片采用免疫组织化学方法对DKK3蛋白水平表达情况进行检测;并对228例肝癌患者预后生存率进行分析。结果 DKK3mRNA水平中在肝癌组织中低表达,在正常组织中高表达(P0.05),DKK3蛋白水平在肝癌组织中低表达(P0.05),DKK3蛋白高表达肝癌患者预后生存时间长于低表达(P0.05),差异有统计学意义。结论 DKK3在肝细胞肝癌组织中低表达,说明DKK3是一种抑癌基因,并且预后结果分析提示DKK3有可能在抑制肿瘤增长,分化,转移中发挥作用。

Expression of DKK3 in hepatocellular carcinoma and its clinical significance

Objective To study the dkk3 expression in hepatocellular carcinoma and its chnical significance. Methods 40 cases of fresh frozen liver carcinoma and paracancerous tissue, 10 patients with normal liver tissue using Real time PCR to detect ex- pression of DKK3mRNA and 228 cases of hepatocellular carcinoma and paracancerous tissue paraffin section were taken using immuno- histochemistry to detect DKK3 protein expression levels; and the survival rate of 228 cases were analyzed. Results Expression level of in liver cancer and high in normal tissues （ P 〈 0. 05 ）, high DKK3 protein expression is low in hepatocellular carcinoma （ P 〈 0. 05 ） , DKK3 protein expression patients with liver cancer survive longer than those with low expression （ P 〈 0. 05 ）, the differences are statis- tically significant. Conclusion DKK3 was low expressed in hepatocellular carcinoma tissue, indicating DKK3 is a tumor suppressor gene; analysis of prognosis prompted DKK3 potentially inhibit tumor growth, differentiation, and metastasis.
