细胞外信号调节蛋白激酶在梗阻性肾组织中的表达及其意义

摘    要:目的 探讨细胞外信号调节蛋白激酶(ERK)在梗阻性肾病的分子发病机制中作用。方法 应用免疫组化方法检测ERK和增殖细胞核抗原(PCNA)在47份梗阻性肾组织中表达,并分析ERK表达与梗阻肾纤维化之间相关关系。结果 梗阻肾的肾小管和间质中ERK表达均高于正常的,但仅间质中两者表达差异有显著性(P〈0.05)。间质中ERK表达与梗阻肾纤雏化程度密切正相关(P〈0.05),而肾小管中两者相关差异无显著性(P〉0.05)。梗阻肾的肾小管和间质中ERK强表达者的PCNA指数均分别显著高于ERK弱表达者(P〈0.01)。结论 ERK活性增强促进细胞增殖增多、从而参与梗阻挂肾病的发病过程,尤其在肾纤维化过程中起着重要作用。

Abstract:[Objective] To investigate the roles of extracellular signal-regulated kinase (ERK) in pathogenesis of chronic obstructive kidney(COK). [Methods] Immunohistochemistry methods were adopted to examine expressions of ERK and proliferation cell nuclear antigen (PCNA) in 47 specimen of COK and 12 specimen of normal kidney. The relationships of ERK expressions and renal fibrosis were analyzed. [Results] ERK expressions in tubular and interstitium of COK were higher than those in normal kidney, respectively, however, statistical difference was only found in interstitium (P <0.05). There was close relationship between ERK expression in interstitium and renal fibrosis of COK (P <0.05), while no significance relationship in tubular (P >0.05). PCNA index of ERK intense expression in tubular and interstitium of COK were significantly higher than those in ERK lower expression (P <0.01), respectively. [Conclusions] Increasing activity of ERK plays an important role in development and progression of COK by promoting cell proliferation, especially in development of renal fibrosis.
