N-乙酰半胱氨酸(NAC)对脂多糖诱导的小鼠感染性早产的影响

摘    要:目的:初步探讨N-乙酰半胱氨酸(N-acetylcysteine,NAC)预防感染性早产的作用机制。方法:采用逆转录-聚合酶链反应(RT-PCR)和免疫组化方法检测脂多糖(lipopolysaccharide,LPS)诱导的感染性早产小鼠模型(模型组)胎盘NF-κBp65、TNF-αmRNA和蛋白的表达,以及NAC干预后(NAC预防组和NAC治疗组)早产率、NF-κBp65和TNF-α的表达变化,并以正常孕鼠作对照。结果:对照组孕鼠无早产的发生,而模型组全部早产,组间比较有显著性差异(P<0.01);对照组NF-κBp65、TNF-α有少量表达,模型组则都显著升高(P<0.05);NAC预防组与模型组比,NF-κBp65和TNF-α表达明显减少(P<0.05),且早产率也明显下降(P<0.05);NAC治疗组与模型组相比无统计学差异。结论:NAC可降低感染性早产小鼠胎盘NF-κBp65和TNF-α的表达,对于感染性早产有保护作用。

Abstract:Objective: To investigate the relationship between NAC and preterm birth. Methods: RT- PCR and immunohistochemistry were used to detect NF-κBp65, TNF-α mRNA and protein expression in the mouse placenta of premature birth, as well as the incidence of preterm birth, NF-κBp65, TNF-α expression changes after the NAC intervention. Results: Without the occurrence of preterm birth in control group, there was significant difference between control group and LPS group (100% preterm birth rate)(P〈0.01). The expression of NF-κBp65 and TNF-α was significantly increased in LPS model group, compared with those in control group(P〈0.05); in the NAC prevention group the expression of NF-κBp65 and TNF-α was decreased and the percentage of preterm birth was also decreased, compared with LPS group (P〈0.05); NAC therapy group had no significant difference compared with LPS group. Conclusion: NAC can reduce the expression of NF-κBp65 and TNF-α in the mouse placenta of preterm birth and has a protective effect of preterm birth.
