摘    要:目的通过研究醇提红曲对去势大鼠骨密度(BMD)、护骨素(OPG)、碱性磷酸酶(ALP)的影响,探讨其防治骨质疏松的机制。方法在乙醇溶解、超声波振荡、水浴氮气条件下蒸去乙醇来提取红曲的有效成分。32只6月龄雌性Wistar大鼠随机分为红曲组、降钙素组、模型组、假手术组。前三组进行去势手术建立骨质疏松模型。术后1周,红曲组、降钙素组、模型组分别予以红曲醇提液灌胃、降钙素皮下注射、生理盐水灌胃,连续给药12周。采用双能X线骨密度仪测量大鼠BMD,采用ELISA测量血清OPG及其他骨代谢指标,分析红曲对大鼠骨代谢影响。结果术后12周红曲组BMD高于模型组,但低于降钙素组及假手术组(均P<0.05);红曲组OPG高于模型组及假手术组,但低于降钙素组(均P<0.000 1);红曲组ALP高于降钙素组及假手术组,但低于模型组(均P<0.000 1)。BMD与血清OPG含量存在正相关关系(r=0.381 67,P=0.031 1),与ALP呈负的直线相关关系(r=-0.660 48,P<0.000 1)。结论红曲可以降低去势大鼠的骨代谢率、减缓骨量减少,推测红曲是通过改变血清OPG含量来改善骨质疏松的。

Abstract:Objective To explore the mechanism of prevention and treatment of osteoporosis by investigating the effect of alcohol extract of monascus on bone mineral density(BMD),osteoprotegerin(OPG),alkaline phosphatase(ALP) in ovariectomized rats.Methods The monascus was extracted after dissolution of ethanol,water bath with nitrogen,and ultrasonic oscillation.Thirty-two 6-month-old female Wistar rats were randomly divided into 4 groups:monascus group,calcitonin group,model group and sham operation group.The rats in first three groups were ovariectomized to induce the model.Then the rats were given intragastric infusion of alcohol extract of monascus in monascus group,subcutaneous injection of calcitonin in calcitonin group,and intragastric infusion of normal saline in model group a week after surgery.The medication continued 12 weeks.The dual energy X ray was used to detect the level of BMD,and ELISA was employed to detect the levels of OPG and other bone metabolism index.Results At the end of 12 weeks,BMD in monascus group was significantly higher than that in model group,but lower than those in calcitonin group and sham operation group(P<0.05).OPG in monascus group was significantly higher than those in model group and sham operation group,but lower than that in calcitonin group(P<0.000 1).ALP in monascus group was significantly higher than those in sham operation group and calcitonin group,but lower than that in model group(P<0.000 1).BMD was positively correlated with the serum content of OPG(r=0.381 67,P=0.031 1),but negatively correlated with ALP(r=-0.660 48,P<0.000 1).Conclusion Monascus can reduce the rate of bone metabolism and slow down the bone loss in ovariectomized rats,which suggests that monascus can improve the osteoporosis by changing the serum content of OPG.
