富血小板血浆对体外培养人脂肪来源干细胞增殖及成脂分化的作用

目的观察富血小板血浆对体外培养的人脂肪来源干细胞增殖与成脂能力的影响。方法用酶消化法获取人脂肪来源干细胞,二次离心法制备自体富血小板血浆,将细胞分为实验组与对照组,实验组以含5%、10%、20%自体富血小板血浆的条件培养液干预,对照组不进行富血小板血浆干预。观察细胞形态学,以XTT比色法测定细胞增殖状况,油红O染色检测细胞成脂活性。结果富血小板血浆实验组较对照组细胞形态饱满、密度增大;XTT比色法显示实验组细胞增殖明显(P<0.01);实验组油红O染色阳性率较对照组增高明显(P<0.01)。结论富血小板血浆对体外培养的人脂肪来源干细胞增殖与成脂分化活性具有显著的促进作用。
