| 摘 要： | 目的区分不同转移能力的鼻咽癌细胞亚群,对于探讨鼻咽癌转移的分子机制、脏器亲和性具有重要的临床和生物学意义。我们拟应用基因芯片技术来分析鼻咽癌细胞株5-8F-EGFP和肝转移亚群5-8F-H3B-EGFP的基因表达谱差异。方法分别提取鼻咽癌细胞株5-8F-EGFP和肝转移亚群5-8F-H3B-EGFP的总RNA,逆转录成荧光标记的cDNA,与Af-fymetrix Human Genome U133A 2.0寡核苷酸芯片杂交（每株重复3次）,用专业的软件和聚类分析法分析基因表达谱。结果得到差异表达基因3767个,其中差异表达2倍以上的基因281个,进一步筛选出16个可能与鼻咽癌肝转移关系密切的基因16个。结论这16个基因能够很好的区分5-8F-EGFP和肝转移亚群5-8F-H3B-EGFP,为进一步寻找鼻咽癌肝转移的临床分子标志物、判断预后缩小了范围。 |
