NcX-ELISA法检测抗ds-DNA在系统性红斑狼疮中的诊断价值

摘    要:
目的探讨双链DNA抗体(抗ds-DNA)的检测方法,寻找1种高特异性、高敏感性的检测方法,为临床诊断及治疗系统性红斑狼疮(SLE)提供可靠的实验依据。方法分别采用间接荧光免疫法(IIFA)、普通ELISA法、放射免疫法(RIA)及NcX-ELISA法,测定50例SLE患者、60例非SLE风湿病患者及30例健康对照者血清中抗ds-DNA。结果采用IIFA法、普通ELISA法、RIA法及NcX-ELISA法测定抗ds-DNA,诊断特异性分别为96.7%、93.3%、95.0%、98.3%,其中以NcX-ELISA法特异性最高,普通ELISA法最低;诊断率分别为71.8%、73.6%、76.4%、86.4%,其中以NcX-ELISA法诊断效率最高,IIFA法最低;检测的敏感性分别为42.0%、50.0%、54.0%、72.0%,以NcX-ELISA法检测敏感性最高,与RIA法相比差异无统计学意义(P>0.05),与普通ELISA法比较差异有统计学意义(P<0.05),与IIFA法比较差异有统计学意义(P<0.005)。结论普通ELISA法特异性低,IIFA法敏感性低,RIA法虽然敏感性、特异性较高,但是容易污染环境;NcX-ELISA法具有高特异性、高敏感性、高诊断率及可批量检测的优点,是检测抗ds-DNA的理想方法。
