大黄麻黄方对大鼠血压及血浆肾素—血管紧张素—醛固酮系统的影响

目的观察大黄麻黄方对大鼠收缩压、肾素活性(PRA)、血管紧张素Ⅱ(AngⅡ)和血管紧张素转换酶(ACE)的时间生物学特征的影响。方法将64只健康雄性SD大鼠随机分为2组,即正常对照组及大黄麻黄方组,每组各32只。正常对照组予蒸馏水10 m L,大黄麻黄方组予大黄麻黄方药液10 m L,均每日2次灌服,连续4周。2组大鼠最后一次灌胃后分别于02:00、08:00、14:00、20:00四个时间点每组分别取8只大鼠,检测各个大鼠收缩压,然后处死大鼠,取心血,采用放射免疫分析法测定血浆PRA、AngⅡ和ACE,然后应用最小二乘法进行余弦拟合法(cosine fitting),分析并比较2组血压及血浆PRA、AngⅡ和ACE的时间生物学特征。结果大黄麻黄方组大鼠服药4周后02:00、08:00、14:00、20:00四个时间点收缩压及血浆PRA、AngⅡ、ACE水平均低于正常对照组同期,比较差异均有统计学意义(P0.05)。与正常对照组相比,大黄麻黄方组大鼠收缩压及血浆AngⅡ、ACE曲线的中值(M)均减小,有统计学意义(P0.05),AngⅡ、收缩压A变化明显,有统计学意义(P0.05),同时PRA、AngⅡ的峰值时间(T)明显变化,有统计学意义(P0.05)。大黄麻黄方组大鼠收缩压、PRA、AngⅡ和ACE曲线的Φ均发生不同程度的位移(P0.05)。结论大黄麻黄方可降低血压,同时对维持血压昼夜节律、改善RASS系统功能有明显影响。

Effects of Dahuang-mahuang formula on blood pressure and plasma renin-angiotensin system in rats

Objective To observe the effects of Dahuang - mahuang formula on the chronobiology character-istic of systolic pressure(SBP),plasma renin activity(PRA),angiotensinⅡ(AngⅡ),angiotensin converting en-zyme(ACE)in rats. Methods 64 healthy male SD rats were randomly divided into two groups that normal control group and Dahuang - mahuang formula group,32 rats in each group. The normal control group received distilled wa-ter(10 mL),the Dahuang - mahuang formula group received Dahuang - mahuang decoction(10 mL),gavage twice daily for four weeks. We measured all the rats′ blood pressure at different times in one day(02:00,08:00,14:00, 20:00)after the last gavage,8 rats in each timing,and the the rats were executed,adopt blood form heart. The plasma PRA,AngⅡ and ACE were measured by radioimmunoassay,and then analyzed the blood pressure its chrono-biology characteristic of PRA,AngⅡ and ACE in two groups by means of osine fitting in two groups. Results The SBP,PRA,AngⅡand ACE after treatment in Dahuang - mahuang formula group were lower than those in control group at four timing(02:00,08:00,14:00,20:00),with statistical differences(P < 0. 05). As compared with normal control group,the M - value of SBP,AngⅡ and ACE in Dahuang - mahuang formula group were decreased, with statistical differences(P < 0. 05),the amplitude(A) of AngⅡand SBP changed obviously(P < 0. 05),and the peak time(T)of PRA and AngⅡchanged obviously,with statistical differences(P < 0. 05). The peak phase(Φ)of SBP,PRA,AngⅡand ACE in Dahuang - mahuang formula group had phase shift in varying degree(P < 0. 05). Con-clusion Dahuang - mahuang formula can decrease blood pressure,and has significant effects of maintain circadian rhythm of blood pressure and improve RASS function.
