150例广州地区汉族人肿瘤坏死因子TNFα-850 C/T基因突变频率的检测

目的了解广州地区汉族人肿瘤坏死因子TNFα-850 C/T基因的分布特点。方法应用PCR-RFLP方法对150例广州地区健康汉族人肿瘤坏死因子TNFα-850 C/T基因进行检测,并结合文献进行了不同地区间的分析比较。结果肿瘤坏死因子TNFα-850 C/C野生型纯合子频率为78.67%,C/T杂合子频率为19.33%,T/T突变型纯合子频率为2.0%,突变等位基因频率为0.1167。结论该方法简便、快速、准确,适合于一般实验室检测及大规模的人群调查,肿瘤坏死因子TNFα-850 C/T基因多态性在不同地区间分布存在着一定的差异。

DETECTION OF MUTATION FREQUENCIES IN THE TNF -850 C/T GENE WITH PCR RFLP IN 150 HEALTH HAN PEOPLE IN GUANGZHOU REGION

Objective To establish a simple, accurate and practical method for TNFct -850C/T genotyping and identify the distribution feature of TNFot - 850C,/T gene in the Hans in Guangzhou region. Methods TNFot - 850C/T gene sequences were amplified by using polymerase chain reaction（PCR）. The PCR products were digestedwith HinclI and subjected to electrophoresis on agarose gels. The patterns of restriction fragment length polymorphism （RFLP） of TNFcL- 850C/T gene were distinguished. Then analysis and comparison among different region were made combined with relevant literatures. Results 150 health individuals were investigated with this PCR - RFLP method, the gene frequency of TNFo~ -850C/T was as followed; TNFa -850 C/C, C/T,T/T genotype frequency： 0. 7867,0.1933,0. 0200, TNFa - 850 C, TNFa - 850 T Genefrequency ： 0. 8833,0. 1167. Conclusion The PCR - RFLP method was considered a rapid and simple technique for obtaining accurate results and also suitable for routine laboratories and large - scale population studies, and TNFct - 850C/T genes distribution in the Hans of different re- gion has some difference.
