rhG-CSF对新生鼠缺氧缺血性脑损伤HIF-1α表达的影响

摘    要:目的:探讨重组人粒细胞集落刺激因子(recombinant human granulocyte colony-stimulating factor,rhG-CSF)对缺氧缺血性脑损伤新生鼠海马区及皮质HIF-1α(hypoxiainducible factor 1α)表达的影响。方法:将7日龄新生Wistar大鼠随机分成假手术组,HIBD组和rhG-CSF治疗组。在乙醚麻醉下行右侧颈总动脉结扎,氮氧混合气缺氧2.5h制备HIBD模型。应用免疫组织化学法检测HIF-1α。结果:免疫组化结果显示:在新生大鼠大脑皮层和海马区,HIBD组较假手术组HIF-1α表达明显增加(P〈0.01),治疗组较HIBD组HIF-1α表达明显增加(P〈0.01)。结论:缺氧缺血性脑损伤后,脑组织HIF-1α表达增加,可能与促进血管形成及抑制神经细胞凋亡有关。rhG-CSF可能通过调节HIF-1α的表达,减少神经细胞凋亡,增强脑组织对缺氧的耐受,从而改善缺氧缺血性脑损伤小鼠的预后。

Abstract:Objective:To investigate the effects of rhG-CSF on the HIF-1a expression in hypoxia ischemia brain damage(HIBD) in neonatal rats.Methods:The Wistar rats,aged 7 days,were divided into Sham-operation group,HIBD group and rhG-CSF group,respectively.The rats of the HIBD group were anesthetized with ethylether.The right common carotid artery was exposed and ligated.Then,they were exposed to hypoxia in a normobaric chamber for 2.5h.The HIF-1αprotein expression was detected by immunohistochemistry.Results:The HIF-1a expression of the HIBD group was increased compared with that of the sham-operation group(P0.01).The HIF-1αexpression of the rhG-CSF group was obviously increased compared with that of the HIBD group(P0.01).Conclusion:The up-regulation expression of HIF-1α may be involved in suppressing apoptosis,and rhG-CSF may enhance the tolerance of the brain tissue to hypoxia and reduce apoptosis possibly by regulating HIF-1αexpression level,suggesting that rhG-CSF can improve prognosis of HIBD rats.
