豆腐果苷磷脂复合物的制备及理化性质研究

摘    要: 目的制备豆腐果苷磷脂复合物并考察其理化性质。方法以豆腐果苷与磷脂的复合率为评价标准,通过正交设计考察反应初始浓度、投料比例和反应时间对复合率的影响;研究豆腐果苷磷脂复合物的理化性质包括晶体特征、相变特征1、H-NMR及溶解性能等。结果确定了豆腐果苷磷脂复合物的最佳制备工艺即在室温下以四氢呋喃为反应溶剂,豆腐果苷质量浓度为5 g·L-1,投料质量比例为1∶5,反应时间为2 h。X-射线衍射分析显示复合物呈现无定型特征;差热扫描显示复合物的相变温度改变;1H-NMR谱图显示豆腐果苷的羟基参与氢键的结合。磷脂复合物明显改善了豆腐果苷在水及正辛醇中的溶解性能。结论确定了制备豆腐果苷磷脂复合物的最佳工艺,并且其磷脂复合物明显的改变了原药的理化性质。

Abstract:OBJECTIVE To prepare hilicid-phospholipid complex and investigate its physic-chemical properties.METHODS The effect of such factors as the concentration of reactants,the ratio of reactants and reactive time on this reaction was investigated using orthogoral design,and the physic-chemical properties of hilicid-phospholipid complex including crystal characteristics,1H-NMR spectra and solubility were examined.RESULTS The best conditions were obtained as follows: the concentration of reactants is 5 g·L-1,the ratio of hilicid to phospholipid is 1∶ 5,and the reactive time is 2 h.X-ray diffraction spectra indicated that the complex exhibits an amorphous characteristic.A change of the transformation temperature of hilicid was showed in DSC spectra.1H-NMR spectra suggested that hydroxy groups of hilicid could interact with phospholipid.Moreover,there is a great improvement in the solubility of hilicid-phospholipid complex both in water and in n-octanol.CONCLUSION The best method to prepare hilicid-phospholipid complex is achieved,and its physic-chemical properties are significantly different from those of hilicid.
