肥胖大鼠模型血浆胆囊收缩素水平的变化及二甲双胍的干预作用

摘    要:目的 :观察高脂饮食诱导的肥胖大鼠血浆胆囊收缩素 (cholecystokinin ,CCK )水平的变化 ,并给予二甲双胍观察其干预效果。方法 :建立高脂饮食诱导的肥胖、胰岛素抵抗大鼠模型 ,检测大鼠体重、空腹胰岛素、空腹血糖及血脂水平。模型建立前及成模后测定大鼠血浆CCK水平变化。并观察给予大鼠二甲双胍灌胃干预治疗后上述指标的变化。结果 :高脂饮食 6个月后 ,大鼠体重、空腹胰岛素及血脂水平较对照组明显增高 (均P <0 0 1) ,血糖增高但无统计学意义 ;肥胖大鼠血浆CCK水平较对照组及入组时显著增高 (均P <0 0 1) ;二甲双胍干预组上述指标明显改善。结论 :肥胖大鼠CCK水平明显增高 ,可能存在类似于胰岛素抵抗的CCK抵抗 ,二甲双胍有明显的干预效果。

Abstract:Objective To investigate the change of the plasma cholecystokinin level in meal-induced fat rats, and examine the int e rference effect of metformin. Methods The fat rat models were developed by given oil-rich food, and the body weight, fasting insulin, fastin g blood sugar and blood lipid of the models were measured. Before and after the e stabishment of the models, the change of plasma cholecystokinin level was determ ined. whilc the intervention effect of metformin was observed. Results The body weight, insulin and lipid of fat rat group were significant elevated than control group after 6 months (all P<0.01), and the plasma ch olecystokinin level increased significantly than before and than control group ( all P<0.01). Those parameters were improved by treatment with metformin. Conclusion The plasma cholecystokinin level increased signifi cantly in fat rats, there maybe the CCK resistance mechanism like insulin resist ance in it, and metformin treatment could improve it.
