吡柔比星联合内皮抑素对治疗人乳腺癌裸鼠移植瘤的研究

摘    要:目的]探讨内皮抑素(E S )联合吡柔比星对人乳腺癌裸鼠移植瘤的肿瘤生长及新生血管生成的抑制作用。方法]构建裸鼠乳腺癌模型,将裸鼠40只,随机分成4组,每组10只。对照组(A组):肿瘤对侧背部皮下注射1×106/0.1 m L正常人成纤维细胞;ES组(B组):肿瘤对侧背部皮下注射1×106/0.1 m L转染了ES的人成纤维细胞。吡柔比星组(C组):尾静脉注射吡柔比星5 mg/kg ,每周1次,共4周,ES+吡柔比星组(D组):肿瘤对侧背部皮下注射1×106/0.1 m L转染了 ES的人成纤维细胞。尾静脉注射吡柔比星5 mg/kg ,每周1次,共4周。ELISA方法检测血清血管内皮生长因子(VEGF),肿瘤组织病理切片观察微血管密度(M VD )。结果]B、C ,D三组抑瘤率分别为61.96%,27.33%和75.16%,D组抑瘤率明显高于其它两组,D组VEGF、MVD与其他各组比较明显减低,且差异有显著性( P<0.05)。结论]ES联合吡柔比星能明显抑制人乳腺癌裸鼠移植瘤生长及新生血管生成,且两者联用具有协同作用。

Study of Pirarubicin Combined with Endostatin for the Treatment of Human Breast Cancer Xen-ografts in Nude Mice

Abstract:Objective]To explore the inhibitory effect of endostatin (ES) combined with pirarubicin(THP) on tumor growth and neoangiogenesis in human breast cancer xenografts in nude mice .Methods]Nude mice models of breast cancer were established .Forty nude mice models were randomly divided into 4 groups with 10 mice in each group .The control group(group A) was given subcutaneous injection of 1 × 106/0 .1mL normal human fibroblasts into the back of contralateral tumor .Group B(group ES) was given subcutaneous injection of 1 × 106/0 .1mL human fibroblast transfected with ES .Group C(pirarubicin group) was given injection of 5mg/kg THP by caudal vein once a week for 4 weeks .Group D(ES+ THP group) was given subcutaneous in-jection of 1 × 106/0 .1mL human fibroblasts into the back of contralateral tumor and injection of 5mg/kg T HP by caudal vein once a week for 4 weeks .ELISA was used to detect serum vascular endothelial growth factor (VEGF) .Pathological section of tumor tissue was used to observe microvessel density (MVD) .Results]Tumor inhibition rate(TIR) of group B ,C and D were 61 .96% ,27 .33% and 75 .16% ,respectively .The TIR of group D was obviously higher than that of other two groups .Compared with other groups ,VEGF and MVD of group D were obviously decreased ,and there was significantly difference( P<0 .05) .Conclusion]ES combined with THP can obviously inhibit tumor growth and neoangiogenesis of human breast cancer xeno-grafts in nude mice ,and has synergetic effect .
