姜黄油对角质形成细胞体外增殖分化影响的研究

目的：探讨姜黄油对角质形成细胞体外增殖分化的影响。方法：以人角质形成细胞株COLO-16细胞为模型，噻唑盐比色法(MTT)检测细胞活性和生长情况；免疫组化SABC法检测姜黄油对增殖细胞核抗原(PCNA)表达的影响；电镜观察姜黄油对细胞超微结构的影响。结果：姜黄油抑制角质形成细胞增殖，随药物浓度增加，其抑制增殖能力增加，PCNA表达逐渐减弱，且对角质形成细胞超微结构有直接损伤作用。结论：姜黄油具有抑制体外角质形成细胞增殖分化的作用，有望成为一种治疗增殖性皮肤病的外用药物。

The effects of turmeric oil on keratinocyte proliferation and differentiation in vitro

Objective: To investigate the effects of turmeric oil on keratinocyte proliferation and differentiation in vitro. Methods: The culture of human transformed keratinocyte cell line COLO-16 was used as a model. A tetrazolium-based colorimetric assay (MTT assay) was used to evaluate cell viability and growth. The effects of turmeric oil on the expression of proliferating cell nuclear antigen (PCNA) were detected by immunohistochemical SABC method. The changes of ultrastructure were observed by transmission electron microscope. Results: Turmeric oil (10 -6 V/V) could inhibit the proliferation of cell line and the inhibiting effect increased as the increase in dosage. PCNA expression decreased gradually with the increasing of concentrations of turmeric oil with a significant difference with controls (P<0.01). The ultrastructure of keratinocytes was directly damaged by turmeric oil. Conclusion: The turmeric oil may inhibit the proliferation and differentiation of keratinocytes in vitro. It is proposed that turmeric oil may be as a new drug for the treatment of proliferating skin diseases.
