纳米级羟基磷灰石梯度涂层对成骨细胞表型因子表达的影响

摘    要:目的研究纳米级羟基磷灰石梯度涂层对成骨细胞表型因子表达的影响。方法对生长于纳米级羟基磷灰石梯度涂层上成骨细胞表达的Ⅰ型前胶原羧基端肽(PICP)、碱性磷酸酶(ALP)和骨谷氨酸蛋白(BGP),于体外培养第1、5、9、13d时进行定量分析,扫描电镜观察细胞形态学改变。以普通级羟基磷灰石涂层和钛合金圆片作为对照。结果纳米级羟基磷灰石梯度涂层组的PICP、ALP和BGP表达均高于对照组,成骨细胞形态好于对照组。结论纳米级羟基磷灰石梯度涂层材料能促进成骨细胞表型因子的表达。

Abstract:Objective To study the effect of nanophase hydroxyapatite biograde-coated plates on osteoblastic phenotype. Methods Osteoblasts were cultured on nanophase hydroxyapatite biograde-coated plates, hydroxyapatite biograde-coated plates and Ti-6Al-4V hydroxyapatite biograde-coated plates, the expressions of carboxyterminal propeptide of type I procollagen (PICP), alkaline phosphatase (ALP) and, bone gla protein (BGP) were assayed at 1, 5, 9 and 13 d. The plates were observed with SEM at the same time. Results The expressions of PICP, ALP and BGP of the cells cultured on nanophase hydroxyapatite material were higher than those of the control group, and the cellular shape was better. Conclusion The expression of osteoblastic phenotype factors was stimulated by nanophase hydroxyapatite biograde-coated material.
