大孔吸附树脂分离纯化单叶铁线莲总皂苷的研究

摘    要:目的研究大孔吸附树脂分离纯化单叶铁线莲总皂苷的工艺条件,为单叶铁线莲总皂苷的工业化生产提供实验依据。方法采用比色法测定总皂苷;比较了5种大孔吸附树脂的静态饱和吸附量、静态解吸率,筛选出适宜的树脂;对最佳树脂进行动态吸附、解吸试验,确定最佳的吸附洗脱工艺。结果 D-101大孔树脂对单叶铁线莲总皂苷有较好的吸附分离性能。最佳工艺条件为:上样液质量浓度为生药0.2 g/mL,pH值为5,上样体积流量为1 BV/h;4 BV蒸馏水洗脱杂质;4 BV 70%乙醇洗脱总皂苷,洗脱体积流量2 BV/h。按此工艺条件生产的总皂苷质量分数达到71.53%,收率86.02%,精制度达238.81%。结论 D-101大孔吸附树脂适合富集纯化单叶铁线莲中总皂苷。
