HINT1基因在喉鳞状细胞癌中的表达和意义

目的组氨酸三聚体核苷结合蛋白1(histidine triad nucleotide-binding protein1,HINT1)基因在喉鳞状细胞癌组织中的表达情况及其与患者临床病理特征的关系,探讨HINT1基因在喉鳞状细胞癌发病过程中的作用。方法通过实时定量PCR和免疫组织化学检测82例喉鳞状细胞癌组织和56例癌旁组织中HINT1mRNA和蛋白水平的表达,分析其蛋白表达与患者临床病理特征的关系。结果喉鳞状细胞癌组织HINT1mRNA水平显著低于癌旁组织(P<0.01),HINT1蛋白表达阳性率显著低于癌旁组织(P<0.05)。晚期喉鳞状细胞癌组织中HINT1蛋白表达显著低于早期喉鳞状细胞癌组织(P<0.05)。结论 HINT1在喉鳞状细胞癌中可能发挥抑癌基因的作用。

Expression of HINT1 gene in laryngeal squamous cell carcinoma and its clinical significance

Objective To investigate the expression of histidine triad nucleotide-binding protein 1() in laryngeal squamous cell carcinoma(LSCC) and its relationship with clinicopathological factors of the patients.Methods 82 cases of LSCC samples and 56 cases of pericancer samples were detected by real time PCR and immunohistochemical staining to analyze mRNA and protein levels,and the relationship between the expression of protein and clinicopathologic factors.Results The expression levels of mRNA in LSCC tissues were downregulated(< 0.01),the positive rate of HINT1 protein expression was significantly lower in LSCC than in pericancer tissues(< 0.05).HINT1 protein expression levels in LSCC tissues were associated with TNM stage of the patients(< 0.05).Conclusion might play a role as a tumor suppressor gene in LSCC.
