乙酰葛根素对 Aβ25-35诱导BV-2小胶质细胞活性的影响

摘    要:目的:研究乙酰葛根素对β淀粉样蛋白( Aβ25-35)诱导的BV-2小胶质细胞活性的影响。方法体外培养BV-2小胶质细胞,用Aβ25-35诱导小胶质细胞建立AD细胞模型,加入不同浓度的乙酰葛根素,显微镜下观察细胞的形态变化;MTT法检测细胞的存活率;ELISA法检测细胞中IL-1β,TNF-α的含量。结果 Aβ诱导后小胶质细胞由分枝状的静息状态转变为阿米巴样的激活状态,而乙酰葛根素可以减轻细胞发生的变化,细胞状态介于空白组与模型组之间;MTT显示乙酰葛根素不会对小胶质细胞的存活率产生影响;ELISA结果显示,与空白组相比,模型组的IL-1β,TNF-α表达水平明显升高,而乙酰葛根素能够降低Aβ诱导IL-1β,TNF-α的表达水平。结论乙酰葛根素能够抑制Aβ25-35诱导的小胶质细胞激活减轻炎性因子的分泌,对阿尔茨海默病具有保护作用。

Abstract:Objective To investigate the effect of Acetylpuerarin on Aβ25-35-induced activation of BV-2 microglia cells.Methods BV-2 microglia cells were cultured in vitro and stimulated with Aβ25-35 to establish the AD model cells .Then the cells were treated with dif-ferent concentration of Acetylpuerarin .The inverted phase contrast microscope was applied to observe the morphological changes of microglia . MTT assay was used to detect the viability of BV-2 cells.ELISA method was used to evaluate the concentration of IL-1βand TNF-αin BV-2 cells.Results Viability of BV-2 cells is not decreased after Acetylpuerarin simulation ,indicating Acetylpuerarin has no toxicity to BV-2 cells.compared with the control group ,the concentration of IL-1βand TNF-αin Aβ25-35 group were significantly increased ,and Acetylpuerarin could down-regulate the expression of IL-1βand TNF-α.Conclusion Acetylpuerarin can inhibit excessive activation of microglia induced by Aβ25-35 and decrease the inflammatory secretion ,it has protective effect on Alzheimer's disease.
