Duchenne肌营养不良模型鼠肌肉内移植小鼠胚胎干细胞后分化潜能的初步研究

摘    要:目的 研究Duchenne肌营养不良模型鼠(mdx鼠)局部肌肉内鼠胚胎干细胞(ESCs)移植治疗后,ESCs的分化、肌肉病理改变及dystrophin蛋白表达变化.方法 eGFP标记的ESCs局部肌肉注射移植入mdx鼠,4周后观察核内移及dystrophin蛋白表达变化.结果 移植后4周mdx鼠核内移改善不明显,dystrophin蛋白阳性表达及肌纤维增加均不明显.结论 局部肌肉内胚胎干细胞移植治疗mdx鼠效果不明显.胚胎干细胞移植后分化为骨骼肌细胞的比例低.

Abstract:Objective To investigate the differentiation of embryonic stem cells(ESCs),the muscular pathological changes and the expression of dystrophin protein after transplantation of mice ESCs into mdx mice skeletal muscle.Methods ESCs labeled with effectively green fluorescence protein(eGFP) were injected into the hindleg gastrocnemius muscle of mdx mice.Four weeks after implantation,differentiation and central nuclei in myofibers and dystrophin protein expression were analyzed.Results Four weeks after implantation,ESCs did not differentiate into muscle cell,and there were no significant changes in the central nuclei in myofibers and dystrophin protein expression in mdx mice.Conclusion The curative effects of ESCs transplantation by local muscular injection for mice with mdx were not satisfactory.The proportion of skeletal muscles differentiated from the transplanted ESCs was low.
