乳腺浸润性导管癌组织中CD147与基质金属蛋白酶-9表达

摘    要:目的探讨CD147与基质金属蛋白酶(metalloprotease,MMP)-9在乳腺浸润性导管癌组织中的表达及意义。方法应用免疫组织化学SP法检测60份乳腺浸润性导管癌石蜡包埋组织中CD147与MMP-9的表达;采用Western blot方法检测28份乳腺浸润性导管癌及癌旁新鲜组织标本中CD147与MMP-9表达情况。结果免疫组织化学结果显示,CD147主要在癌细胞的胞膜及胞质中表达,在间质中不表达;MMP-9主要在癌细胞的胞质中表达,在间质中不表达;CD147与MMP-9表达在组织学分级及淋巴结转移上差异有统计学意义(P〈0.05);在年龄、肿块大小及临床分期上差异无统计学意义(P〉0.05);2种蛋白在乳腺浸润性导管癌组织中表达有明显相关性(r=0.769,P〈0.05)。Western blot结果显示,乳腺浸润性导管癌组织中CD147与MMP-9表达高于癌旁乳腺组织(P〈0.05),且有淋巴结转移者高于无淋巴结转移者(P〈0.05)。结论 CD147与MMP-9在乳腺浸润性导管癌组织中表达明显增高,可能在乳腺癌发生、发展中发挥重要作用。

Abstract:Objective To study the expressions of CD147 and matrix metalloprotease-9 in breast infiltrating duct carcinoma.Methods The expressions of CD147 and matrix metalloprotease-9 were examined in 60 cases of paraffin-embedded invasive ductal carcinoma tissue with immunohistochemcal staining SP method and 28 cases of invasive ductal carcinoma and adjacent freshly-taken tissues with Western blot.Results Immunohistochemistry showed that CD147 was mainly expressed in the membrane and the cytoplasm of cancer cells,and not expressed in the stroma.Matrix metalloprotease-9 was mainly expressed in the cytoplasm of cancer cells,and not expressed in the stroma.The expressions of CD147 and matrix metalloprotease-9 were correlated with histological grade and lymphatic metastasis(P0.05),and not correlated with age,tumor size and clinical stage(P0.05).The expression of two proteins had a significant correlation in invasive ductal carcinoma(r=0.769,P0.05).Western blot results showed that the expressions of two proteins were significantly higher in breast cancer than those in adjacent normal tissues(P0.05),and higher in lymphatic metastasis than in those without lymph node metastasis(P0.05).Conclusion The levels of CD147 and matrix metalloprotease-9 proteins in breast invasive ductal carcinoma increase significantly,which plays an important role in the development and progression of breast cancer.
